Impacto de la maquinaria de metilacion de adenosinas (m6A) en la expresion genica de VIH-1

N6-metiladenosina (m6A) es la modificación interna más abundante en los ARNm eucariontes, con un importante impacto
en diversas etapas de la expresión génica. Esta modificación es dinámica y se encuentra altamente regulada por proteínas
escritoras, borradoras y lectoras. El complejo encargado de metilar las adenosinas del ARNm (escritoras), esta compuesto por
las metiltransferasas METTL3 y METTL14 junto con WTAP. Las proteínas borradoras corresponden a las demetilasas FTO y
ALKBH5, que remueven específicamente la metilación confiriendo reversibilidad al proceso. Las proteínas lectoras detectan
e interpretan m6A, y corresponden principalmente a miembros de la familia YTH (YTHDF1, YTHDF2 e YTHDF3). Se ha visto
que la interacción de YTHDF1 con sus ARNm blanco favorece la traducción mientras que la unión de YTHDF2 promueve la
degradación. Diversos análisis transcriptómicos han revelado la presencia de m6A en una gran cantidad de ARNm celulares
y virales incluyendo el ARNm completo de VIH-1, sugiriendo una fina regulación de la expresión génica viral a través de la
modificación química de este transcrito. En este trabajo analizamos el impacto de los diferentes componentes de la maquinaria
de m6A en la expresión del ARNm completo de VIH-1. Nuestros resultados indican que la expresión de la poliproteína Gag
se beneficia de la presencia de m6A en el ARNm completo. Interesantemente, encontramos que pese a estar involucrada en
la degradación de los ARNm, la expresión ectópica de YTHDF2 estimula la expresión del ARNm completo, sugiriendo que el
reconocimiento de m6A por esta proteína lectora modula positivamente la expresión génica viral. Sin embargo, la expresión
ectópica de un mutante de YTHDF2 incapaz de unir m6A ejerce el mismo efecto que la proteína silvestre, indicando un efecto
independiente de m6A. Análisis informáticos indican que el dominio N-terminal de YTHDF2 es intrínsecamente desordenado
y posee varios sitios de unión a ARN no-canónicos previamente descritos en otras proteínas. Estos resultados indican que
la expresión del ARNm completo de VIH-1 se favorece de la maquinaría celular de m6A. Nuestros datos también muestran
por primera vez un efecto de YTHDF2 independiente de m6A que podría estar asociado a sitios de unión a ARN no-canónicos.
