Existe una vía alternativa asociada a la diferenciación de células B que es activada por daño al ADN. Esta activación resulta en la fosforilación, inactivación y translocación al citoplasma del co-activador transcripcional CRTC2, lo que resulta en la down-regulación de un set de genes asociados a proliferación. Resultados preliminares obtenidos por nuestro laboratorio detectaron la interacción de esta proteína con el co-represor transcripcional mSIN3A y la histona de-acetilasa 1 (HDAC1) en una línea de células B (Ramos) y un enriquecimiento de mSIN3A en la región promotora de genes blanco de CRTC2. Considerando esta información nos planteamos la siguiente hipótesis: El complejo mSIN3A/HDAC1 participa en la regulación transcripcional de genes blanco de CRTC2 en células B del centro germinal durante su diferenciación inducida por quiebres al ADN. Para responder esta hipótesis establecimos una serie de objetivos con distintas aproximaciones experimentales. A través de ensayos de Western blot e inmunoprecipitación pudimos no sólo detectar la presencia de estas proteínas, sino que establecer interacciones entre éstas. mSIN3A interacciona con HDAC1 y proteínas del complejo SWI/SNF como Brg1 y Brm, y además interacciona con MeCP2 en células control y bajo estrés genotóxico. Utilizando ensayos de inmunoprecipitación de cromatina pudimos determinar que bajo estrés genotóxico existe un incremento significativo de la localización de mSIN3A y HDAC1 en la región promotora de los genes blanco de CRTC2. Estos resultados se correlacionan con cambios en el perfil de marcas epigenéticas de activación y represión transcripcional analizadas. Existe una disminución en el enriquecimiento de las marcas de activación H3K27Ac y H3K9Ac y un incremento en la marca de represión H3K9me3. Al analizar la importancia de estas proteínas en la regulación transcripcional de estos genes, no pudimos detectar una clara contribución de ellas. A través del uso de sh-RNA y posteriormente inhibidores, detectamos que tanto las aproximaciones metodológicas como el estrés genotóxico contribuyen en la down-regulación de estos genes, posiblemente a través de mecanismos diferentes. Es posible que la activación de vías apoptóticas y un arresto del ciclo celular producto de la transducción lentiviral, y la down-regulación de HDAC1, contribuyan de forma significativa en la down-regulación de los genes de CRTC2 analizados. Es necesario utilizar metodologías adicionales y evaluar marcadores de muerte y ciclo celular con el fin de elucidar los mecanismos implicados en esta regulación. There is an alternative pathway associated with differentiation of B cells that is activated by DNA damage. This activation results in the phosphorylation, inactivation and translocation to the cytoplasm of the transcriptional co-activator CRTC2, which results in down-regulation of a set of genes associated with proliferation. Preliminary results obtained by our laboratory detected the interaction of this protein with the transcriptional co-repressor mSIN3A and the histone de-acetylase 1 (HDAC1) in a B cell line (Ramos) and an enrichment of mSIN3A in the promoter region of CRTC2 target genes. Considering this information, we propose the following hypothesis: The mSIN3A / HDAC1 complex participates in the transcriptional regulation of CRTC2 target genes in B cells of the germinal center during their differentiation induced by DNA breaks. To answer this hypothesis, we established a series of objectives with different experimental approaches. Through Western blot assays and immunoprecipitation, we were able not only to detect the presence of these proteins, but to establish interactions between them. mSIN3A interacts with HDAC1 and SWI / SNF complex proteins such as Brg1 and Brm and interacts with MeCP2 in control cells and under genotoxic stress. Using chromatin immunoprecipitation assays, we were able to determine that under genotoxic stress there is a significant increase in the localization of mSIN3A and HDAC1 in the promoter region of the CRTC2 target genes. These results correlate with changes in the profile of epigenetic marks of activation and transcriptional repression analyzed. There is a decrease in the enrichment of the activation marks H3K27Ac and H3K9Ac and an increase in the repression mark H3K9me3. When analyzing the importance of these proteins in the transcriptional regulation of these genes, we could not detect a clear contribution of them. Through shRNA and inhibitors, we detect that both methodological approaches and genotoxic stress contribute to down-regulation of these genes, possibly through different mechanisms. It is possible that the activation of apoptotic pathways and cell cycle arrest product of the lentiviral infection and the down-regulation of HDAC1, respectively, contribute significantly to the down-regulation of the CRTC2 genes analyzed. It is necessary to use additional methodologies and evaluate markers of death and cell cycle to elucidate the mechanisms involved in this regulation.
