Filifolinol es un compuesto de origen natural que se encuentra presente en el exudado resinoso de Heliotropium filifolium, un arbusto endémico de Chile, que crece en la región de Atacama. Estudios biológicos de filifolinol y algunos de sus derivados naturales y semisintéticos sobre patógenos que afectan la industria acuícola y estudios in vivo en Salmón del Atlántico, han mostrado un gran potencial para su uso veterinario con actividades tales como: antiviral, antibacteriano e inmunoestimulante. Uno de los mayores problemas que afectan a la industria acuícola chilena, la cual representa alrededor del 30% de la producción mundial, es la infección de los cultivos y ecosistemas circundantes. Además, la contaminación por el uso excesivo de antibióticos poco eficientes para el tratamiento de los patógenos hace necesaria la búsqueda de nuevos tratamientos más eficaces y menos tóxicos. Basados en estos antecedentes, este trabajo se centra en la síntesis total de algunos derivados de filifolinol. La estructura de filifolinol es clasificada como un derivado aromático geranilado conformado por una estructura de dihidrobenzofurano sustituido enlazado a través de un carbono de tipo espiro a un ciclohexano sustituido y que presenta tres centros estereogénicos en su estructura. La ruta retrosintética planteada es de tipo convergente. Por una parte, se sintetizó el fragmento cíclico a partir de compuestos accesibles comercialmente y por otra, se sustituyó el fragmento aromático para la posterior convergencia y obtención de las estructuras finales a través de una anillación. Para la síntesis del cicloalcano se utilizó 3-pentanona y acrilato de metilo además de herramientas de síntesis clásicas como: reacciones de tipo Michael, adiciones a carbonilo, metilaciones y reducción con agentes clásicos, entre otras. La reducción regioselectiva del grupo carbonilo se logró utilizando borohidruro de sodio en cantidades estequiométricas. Para la convergencia del sistema aromático y el sistema alifático, se realizó una adición nucleofílica sobre el grupo carbonilo del sistema alifático. Se formó in situ el derivado organolitiado del sistema aromático utilizando litio metálico o n-butillitio, permitiendo la formación del enlace C-C. En el último paso de la ruta sintética, se utilizó una reacción de activación Csp2-H catalizada por paladio, para la anillación a través de la formación de un enlace C-O a partir de un grupo hidroxilo enlazado a Csp3. El primer uso de esta metodología sintética fue reportado en 2010 y hasta la fecha existen pocos trabajos publicados que realicen este tipo de conexiones utilizadas en la síntesis total de productos naturales. El presente trabajo muestra su novedad al utilizar esta reacción para la obtención del carbono espiránico de la estructura de análogos de filifolinol a través de una anillación intramolecular. Finalmente, se logró sintetizar dos nuevos análogos de filifolinol, los cuales no presentan la misma configuración en los centros estereogénicos correspondientes con la estructura de filifolinol. Este resultado no es sorprendente ya que la síntesis se realizó sin la utilización de auxiliares o reactivos quirales. Sin embargo, el trabajo aquí expuesto propone un interesante punto de partida para la obtención de este compuesto natural. Filifolinol is a naturally occurring compound present in the resinous exudate of Heliotropium filifolium, a Chilean endemic shrub that grows in the Atacama region. Biological studies of filifolinol and some of its natural and semisynthetic derivatives against pathogens that affect the aquaculture industry and in vivo studies in Atlantic salmon have shown a great potential for its veterinary use, such as an: antiviral, antibacterial and immunostimulant. One of the biggest problems affecting the Chilean aquaculture industry, which represents around 30% of the global production, is the infection of the ecosystems and salmon farms. Moreover, the contamination due to the excessive use of inefficient antibiotics represent another problem. Thus, new and less toxic treatments against these pathogens are necessary. Based on this, the current work is focused on the total synthesis of filifolinol derivatives. The structure of filifolinol is classified as an aromatic geranyl derivative consisting of a dihydrobenzofuran structure linked through a spiro carbon to a substituted cyclohexane with three stereogenic centers in its structure. The proposed retrosynthetic route was convergent. On one hand, the cyclic fragment was synthesized from commercially accessible compounds and on the other hand, the aromatic fragment was substituted for the subsequent convergence and obtention of the final structures through an annulation. For the cycloalkane synthesis, 3-pentanone and methyl acrylate were used besides classic synthetic tools such as: Michael reaction, carbonyl additions, methylations and reduction with classic agents, among others. A regioselective reduction of the carbonyl group succeeded using sodium borohydride in stoichiometric quantities. For the convergence of the aromatic and aliphatic systems, a nucleophilic addition was done on the carbonyl group of the aliphatic system. The corresponding organolithium reagent of the aromatic system was formed in situ by using metallic lithium or n-butyllithium, allowing the formation of the C-C bond. In the last step of the synthetic route, a palladium catalyzed Csp2-H activation reaction was used for the annulation through the formation of the C-O bond from a hydroxyl group linked to a Csp3. The first use of this synthetic methodology was reported in 2010 and only few reports describing this kind of methodologies applied to the synthesis of natural products have been published. This work shows its novelty by using this reaction for the formation of the spiro carbon in the structure of the filifolinol derivatives through an intramolecular annulation. Finally, two new filifolinol derivatives could be synthesized with different configurations of the stereogenic centers regarding the filifolinol structure. This is not surprising because the synthesis was performed without the use of chiral auxiliaries or reagents.
