| dc.description | Histiocytoma, mastocytoma and transmissible venereal tumors are frequent in dogs. The differential diagnostic between these tumors is important, mainly when the transmissible venereal tumor is ectopic. Image cytometry can be used as an auxiliary tool in the differential diagnostic of the histological similar pathology, such as these tumors. The objective of this work was to characterize the nucleaus and the chromatin morphometry of the neoplasic cells of these tumor types and to evaluate the potential of this methodology as differential diagnostic of these pathologies. Fifteen histological slides of each type of the tumor were used and one hundred nucleus of each case were analyzed. Geometric basic characteristics (area, perimeter and ellipticity) traditionally used as auxiliary tools in the histopathological diagnostic, as well as more elaborated geometric measures and the characterization of the nuclear texture through of lacunarity and others methods considered in this work were used Variance and canonical discriminant analysis were made for statistic evaluation. The morphometry using basic measurements was inefficient to identify correctly these tumor types. However, this methodology showed to be a good tool for differential diagnosis of the histiocytoma and transmissible venereal tumor. | dc.description | Histiocitoma, mastocitoma e tumor venéreo transmissível são tumores frequentes em cães. É importante o diagnóstico diferencial entre estes tumores, principalmente quando o tumor venéreo transmissível é ectópico. A citometria de imagem pode ser usada como auxiliar no diagnóstico diferencial de patologias semelhantes histologicamente, como nos casos destes tumores. O objetivo deste trabalho foi caracterizar morfometricamente o núcleo e a cromatina das células neoplásicas destes tumores e avaliar o potencial deste tipo de análise como ferramenta auxiliar no diagnóstico diferencial destas patologias. Foram utilizados 15 cortes histológicos de cada tipo de tumor, sendo analisados 100 núcleos de cada caso. Além das características geométricas básicas (área, perímetro e elipsidade), tradicionalmente utilizadas como auxiliar no diagnóstico histopatológico, também foram utilizadas medidas geométricas mais elaboradas, bem como a caracterização da textura nuclear através de métodos tradicionais, como a lacunaridade e através de novos métodos propostos neste trabalho. Realizou-se análise de variância e de discriminantes canônicas para a avaliação estatística. A morfometria utilizando medidas básicas mostrou-se ineficiente para diferenciar estes tumores. Com o uso das demais medidas otimizadas pela análise de discriminante canônica, esta metodologia revelou-se uma excelente ferramenta eficiente para o diagnóstico diferencial de histiocitoma e tumor venéreo transmissível. Para o diagnóstico diferencial dos três tipos de tumores esta metodologia deve ser usada somente como ferramenta auxiliar. | |
