dc.description | Here we report a genome-wide association study for non-pathological pinna morphology in over 5,000 Latin Americans. We find genome-wide significant association at seven genomic regions affecting: lobe size and attachment, folding of antihelix, helix rolling, ear protrusion and antitragus size (linear regression P values 2 x 10(-8) to 3 x 10(-14)). Four traits are associated with a functional variant in the Ectodysplasin A receptor (EDAR) gene, a key regulator of embryonic skin appendage development. We confirm expression of Edar in the developing mouse ear and that Edar-deficient mice have an abnormally shaped pinna. Two traits are associated with SNPs in a region overlapping the T-Box Protein 15 (TBX15) gene, a major determinant of mouse skeletal development. Strongest association in this region is observed for SNP rs17023457 located in an evolutionarily conserved binding site for the transcription factor Cartilage paired-class homeoprotein 1 (CART1), and we confirm that rs17023457 alters in vitro binding of CART1. | |
dc.description | Aquí informamos un estudio de asociación de todo el genoma para la morfología del pabellón auricular no patológico en más de 5000 latinoamericanos. Encontramos una asociación significativa en todo el genoma en siete regiones genómicas que afectan: el tamaño y la unión del lóbulo, el plegamiento del antihélix, el giro del hélix, la protrusión de la oreja y el tamaño del antitrago (valores P de regresión lineal de 2 x 10 (-8) a 3 x 10 (-14) ). Cuatro rasgos están asociados con una variante funcional en el gen del receptor de ectodisplasina A (EDAR), un regulador clave del desarrollo del apéndice de la piel embrionaria. Confirmamos la expresión de Edar en la oreja del ratón en desarrollo y que los ratones deficientes en Edar tienen un pabellón auricular de forma anormal. Dos rasgos están asociados con los SNP en una región que se superpone al gen T-Box Protein 15 (TBX15), un determinante importante del desarrollo esquelético del ratón. La asociación más fuerte en esta región se observa para SNP rs17023457 ubicado en un sitio de unión conservado evolutivamente para el factor de transcripción Homeoproteína 1 de clase emparejada de cartílago (CART1), y confirmamos que rs17023457 altera la unión in vitro de CART1. | |
