dc.description | Aims:A new L-asparaginase produced byStreptomyces ansochromogenesUFPEDA 3420 actinobacteria was used in this study against humanlymphocyte cultures to evaluate the immunological profile induced by thisenzyme.Methods and Results:Cultures of lymphocytes were stimulated withS.ansochromogenesL-asparaginase, and cytotoxicity, cell viability, cell stimulationand cytokine production were analysed. This newS. ansochromogenesL-asparaginase induced activation and proliferation of the TCD8+lymphocytesubset and produced higher TNF-a, IFN-c, IL-2 and IL-10 levels in a 24-hassay.Conclusion:Streptomyces ansochromogenesL-asparaginase is a promisingmolecule to be used inin vivomodels and to deepen preclinical tests againstacute lymphoblast leukaemia.Significance and Impact of Study:L-asparaginase is an indispensablecomponent of the chemotherapeutic treatment of acute lymphoblast leukaemia(ALL) and acute myeloid leukaemia (AML). Currently, drugs such asAsparaginaseâ, Kidrolaseâ, and Elsparâand Erwinaseâare efficient againstleukemic disease, but promote immunosuppression and other side effects inhuman organisms. Our purifiedS. ansochromogenesL-asparaginase showedpromissory results inducing,in vitro, higher immunostimulation in humanPBMC, especially in T CD8+lymphocyte subsets. | |
