This research evaluated the effect of Resveratrol (RES) on the cryopreservation of bull
spermatozoa by conventional slow freezing or vitrification and its impact on sperm quality. For
that, twenty-four bull ejaculates were collected using an artificial vagina, diluted with TCG-EY
(tris, citric acid, and glucose + 6% egg yolk) medium, and then four groups were conformed
according to the cryopreservation method (conventional freezing [CF] and vitrification [VIT])
and RES supplementation (0.05 mM and 0 [CONTROL]): CF-RES, CF-CONTROL, VIT-RES,
and VIT-CONTROL. Freezing process was performed using 5% glycerol, 0.25 mL straws, and
exposing to liquid nitrogen (NL2) vapors. Vitrification was performed using 100 mM sucrose and
submerging the samples in pellets (30 µL) into NL2. Overall, CF produced a better sperm quality
cryoresponse than VIT procedure, regardless of RES addition supplementation. RES improved
(P<0.05) the sperm membrane integrity before freezing and the total motility before vitrification
compared to their controls. After freezing-thawing, RES increased (P<0.05) total and
progressive motility, as well as plasmatic and acrosomal membrane integrity (CF-RES vs. CF CONTROL). After vitrifying-warming, RES increased (P<0.05) the beat cross frequency (BCF)
and decreased (P<0.05) oxidative stress (VIT-RES vs. VIT-CONTROL). In conclusion,
Resveratrol improved the motility and plasmatic and acrosomal membrane integrity of bull
sperm before and after freezing and reduced oxidative stress after vitrification. Further research
is recommended to evaluate the effect of Resveratrol on fertility.
