Actividad antiproliferativa y genotóxica de aceites esenciales de Salvia sagittata “salvia azul” y Minthostachys mollis “muña” provenientes de Cachicadán, Región La Libertad

Resumen
El uso de plantas medicinales en la población peruana cada día se evidencia como primera elección para menguar sus dolencias. En tal sentido, la presente investigación tuvo como objetivo determinar la actividad antiproliferativa y genotóxica de los aceites esenciales de Salvia sagittata “salvia azul” y Minthostachys mollis “muña” provenientes de Cachicadán. La evaluación de la actividad antiproliferativa se realizó en células tumorales de MCF-7, NCI-ADR / RES, NCI-H460, PC-3 y una línea no tumoral HaCaT. El efecto genotóxico se analizaron en células meristemáticas de 27 bulbos de Allium cepa. Se dispuso 9 grupos de trabajo: control negativo, control positivo, DMSO y tratamientos de ambos aceites (3, 30 y 300 ug/mL) por una exposición total de 24 horas. Adicionalmente, se determinó la caracterización química de los aceites esenciales por cromatografía de gases/espectrometría de masas. Los aceites esenciales no presentan actividad antiproliferativa, de acuerdo con los criterios descritos por Fouche et al tiene un TGI>150 siendo inactivo. Se observaron diferentes aberraciones cromosómicas en los aceites esenciales evaluados tales como: puentes en anafase, cromosomas fragmentados, cromosomas pegajosos, C-metafase, binucleares, núcleos desplazados y micronúcleos. Los principales compuestos de los aceites esenciales fueron: dihydro carvone <cis-> (18,95%), carvone (18,86%), piperitone epoxide <cis-> (10,59%), carvacrol acetate (9,66%), carvacrol (7,37%) correspondientes para Minthostachys mollis; por otro lado, Salvia sagittata presentó: Bicyclogermacrene (12,16%), Linalyl anthranilate (11,11%), Caryophyllene (10,01%). Los aceites esenciales de Salvia sagittata y Minthostachys mollis no tienen actividad antiproliferativa y se le podría atribuir un efecto genotóxico. The use of medicinal plants in the Peruvian population every day is evidenced as the first choice to reduce their ailments. In this sense, the present investigation aimed to determine the antiproliferative and genotoxic activity of the essential oils of Salvia sagittata "blue sage" and Minthostachys mollis "muña" from Cachicadán. Evaluation of antiproliferative activity was performed on tumor cells of MCF-7, NCI-ADR/RES, NCI-H460, PC-3, and a non-tumor HaCaT line. The genotoxic effect was analyzed in meristematic cells of 27 bulbs of Allium cepa. 9 working groups were arranged: negative control, positive control, DMSO and treatments of both oils (3, 30 and 300 ug/mL) for a total exposure of 24 hours. Additionally, the chemical characterization of the essential oils was determined by gas chromatography/mass spectrometry. Essential oils don´t present antiproliferative activity, according to the criteria described by Fouche et al, they have a TGI>150 being inactive. Different chromosomal aberrations were observed in the essential oils evaluated, such as: anaphase bridges, fragmented chromosomes, sticky chromosomes, C-metaphase, binuclear, displaced nuclei and micronuclei. The main compounds of the essential oils were: dihydro carvone <cis-> (18,95%), carvone (18,86%), piperitone epoxide <cis-> (10,59%), carvacrol acetate (9,66%), carvacrol (7,37%) corresponding to Minthostachys mollis; on the other hand, Salvia sagittata presented: Bicyclogermacrene (12,16%), Linalyl anthranilate (11,11%), Caryophyllene (10,01%). The essential oils of Salvia sagittata and Minthostachys mollis do not have antiproliferative activity and a genotoxic effect could be attributed to them.
