dc.description.abstract | La glicina actúa regulando la vía TNF-α/NF-ĸB bloqueando los procesos inflamatorios en adipocitos, a través de la participación de un receptor especifico, asociado a canales de Cl-, compuesto de tres subunidades distintas α, ᵦ y gefirina. Actualmente no existen estructuras cristalográficas de las isoformas α1, α2 y α4, solamente de α3 de humano, por lo que esta subunidad fue utilizada para obtener los modelos por homología para el resto de las isoformas, permitiendo determinar la energía de unión (ΔGu) entre las isoformas del GlyR/glicina. Los resultados indicaron que la unión está conducida por interacciones electrostáticas en las diferentes isoformas, principalmente por la participación de un glutámico y treonina en el sitio de unión del GlyR/glicina. La α3 presentó mayor interacción, ya que su expresión es abundante en las células de médula. Estos resultados comprueban que si existe interacción entre la glicina y su receptor. | |
dc.description.abstract | Glycine acts regulating TNF-α/NF-ĸB pathway, blocking inflammatory processes in adipocytes, through the participation of a specific receptor, associated with Cl- channels, composed of three different α, ᵦ and gephyrin subunits. Currently there are no crystallographic structures of the α1, α2 and α4 isoforms, only of human α3, therefore, this subunit was used to obtain homology models for the rest of the isoforms, allowing the determination of the binding energy (DGb) between the GlyR/glycine isoforms. The results indicated that the binding is driven by electrostatic interactions in the different isoforms, mainly by the participation of a glutamic and threonine in the GlyR/glycine binding site. α3 showed a greater interaction since its expression is abundant in marrow cells. These results prove that there is an interaction between glycine and its receptor. | |
