Propolis is a resinous substance obtained by bees by resins collection in tree bark, plant sprouts and other parts of the plant tissue and its main function the protection and asepsis of the hive. Its composition depends on the area where vegetation is encountered, resulting in many different types of propolis. Recently, propolis is the object of many studies because of their biological properties such as antibacterial and antioxidant, mainly due to the presence of phenolic compounds. For identification and quantification of these phenolic, the ethanol extract of raw propolis (EEP), produced by selected bees, was submitted to chromatographic analysis by liquid chromatography of high efficiency with diode array detector (HPLC-DAD). From the chromatogram, it was concluded that the EEP has a complex chemical profile and by comparison with standards, it was possible to identify the caffeic acid, coumaric and ferulic quantified the concentrations of 14.33 mg, 37.83 mg and 18,31mg per 100 g of propolis, respectively. In order to concentrate the phenolic compounds EEP was subjected to solid phase extraction (SPE) and also analyzed by HPLC, and only the ferulic acid concentration was identified with 10.58 mg per 100 g of propolis. The analysis provided a more efficient EEP chemical characteristics compared with the extract obtained after purification utlizando SPE technique. To ensure reliability of the chromatographic method, the sensitivity parameters, limits of detection and quantification, precision and linearity were calculated.
