Resumen
Due to the resistance of Helicobacter pylori to antibiotics used in the conventional treatment of gastroduodenal pathologies, several researches have been developed currently focusing on the eradication of this bacterium. Among the possible alternative therapies, those that use cinnamic acid, a compound that can act on the defense mechanism of H. pylori by inhibiting the urease enzyme, stand out. In this context, the present work evaluated the pharmaceutical potential of cinnamic and 4-methoxycinnamic acids and their corresponding zinc complexes, zinc cinnamate [Zn(CI)2] and zinc 4-methoxycinnamate [Zn(MCI)2]. For this purpose, the pharmacokinetic and pharmacodynamic properties of the mentioned compounds were evaluated, through the use of several computational tools, as well as the in vitro inhibition potential of the urease enzyme. Regarding the pharmaceutical form, the use of a polymeric encapsulation system, based on chitosan polymer, was evaluated for the controlled release of bioactive substances at gastric pH. The pharmacokinetic properties indicated that the compounds have ADME properties consistent with bioactive drug candidates; the urease enzyme inhibition potential was confirmed by means of in silico and in vitro evaluations and the encapsulation and controlled release potential was confirmed by the use of gastric simulation fluid. In addition, the compounds were confirmed are not cytogenic and genotoxic. The results presented indicate that the four substances evaluated have the potential to be used as drugs for the eradication of H. pylori.
