Resumen
Crinum kunthianum is an endemic species of Ecuador, belonging to the family, Amaryllidaceous, which is characterized by alkaloids, the importance of the treatment of Alzheimer's disease. The objective of the present titration work is to determine the inhibitory activity of the alkaloidal extract of Crinum kunthianum bulbs on the enzymes acetylcholinesterase and butyrylcholinesterase. The alkaloidal of this species were obtained through a protocol of pH changes and the use of organic solvents, obtaining a total alkaloid yield of 1,899 %. Identification of the alkaloids present in the purified extract by gas chromatography coupled to Mass Spectrometry. The determination of the inhibitory activity of acetylcholinesterase and butyrylcholinesterase was carried out by means of a colorimetric method, using galantamine as the reference compound, calculating the inhibitory concentration 50 (IC50). A total of 11 alkaloids were identified, bowdensin being the most abundant with 63.0587 (μg gal / 100mg DW), followed by liquorine with 35.3075 (μg gal / 100mg DW) and in less cases the anhydrolicorin alkaloids are found, 1-0 -acetylcillin and 2-0-acetylcillin. The inhibitory activity of the alkaline extract of Crinum kunthianum on the acetylcholinesterase enzyme was evaluated by varying the concentration of the cosolvent, the best result being dilution with dilution 2 of cosolvent B whose IC50 was 44.26 ± 0.64, compared to IC50 0.49 ± 0.20 corresponding to the galantamine reference compound. Regarding the inhibitory activity on butyrylcholinesterase it is said that in the dilution with dilution 3 of cosolvent B an IC50 of 39.14 ± 2.03 was obtained, while galantamine gave an IC50 result of 15.36 ± 0.98. The inhibitory activity of the primary science on the enzymes is considered high although the activity of galantamine is higher. It is recommended to carry out more studies on Crinum kunthianum, since it has some metabolites of pharmacological interest and there is very little information about this species.
