Theemergenceofinfectionscaused bymulti-drug resistantbacteriaworldwideand thesubstantialincreasein cancerhasled tothesearchforinnovativetherapies.Inthisway,theuseofresearcheswithdrugsalreadyinclinicalusethatcould providemoreeffectivetreatmentalternativesto overcomethesecurrentchallenges,dueto thelimited numberofavailabletherapeuticagents,characterizing therepositioning orredirectionofdrugs,wasused.Amitriptylineisatricyclicantidepressantnotwidelyused intheclinicforthispurposebecauseofitsmarked sideeffectsinthedoseat whichithasto beused.However,atalowerdose,itsuseiscurrentlyinthetreatmentofpain,suchasprophylacticmigraineand cancerpain.However,studieshavedemonstrated theiractivityagainstvarioustypesofbacteriaand fungi.Duetoitsdiversified biologicalactivitycoupled withtherelativelylow priceand theisolation ofmultiresistantbacteria(MDR)mainlyatthehospitallevel,wewereencouraged to conductthisresearch.Oftendrugsthat arecapableofcleaving DNAdouble-stranded haveboth antibacterialand anti-cancereffects.Thisknowledgeand theexistenceofonlyhypothesesin theclarificationofthemechanismofactionofthisdrugistheobjectiveofthisstudyinwhich weintend to evaluatetheinvitrobiologicalactivityofthedrugamitriptylineagainstAmerican Standard CultureCollection (ATCC)referencestandard strainsand clinicalisolatesMDR,bydetermining theminimuminhibitoryconcentration(MIC),minimumbacterialconcentration (MBC),tolerancelevel,fractionalinhibitoryconcentrationindex(FICI),associated ornot with antibioticsused inclinicalpracticeand cleavageabilityofthepUC18 doublestranded plasmidDNA.
