Resumen
Inactivation of tumor suppressor genes, whose products exert an inhibitory influence on cell cycle progression, can lead to neoplastic transformation. In acute myeloid leukemia (AML), the frequency of p53 gene mutations ranges from 4 to 15% in populations from USA and Europe. In an attempt to investigate the frequency of point mutations in the p53 gene in AML Brazilian patients, DNA samples of 35 patients were studied using PCR-SSCP techniques, screening exons 4-10. Mutations were identified in bone marrow DNA in 5 of the 35 AML patients (14.3%), a frequency similar to those reported for Northern American and European populations. The overall survival of patients with mutations in the p53 gene was significantly shorter than for patients without mutations. 7 1 13 19 Diamandis, E.P., "Clinical applications of the p53 tumor suppressor gene" (1995) Clinica Chimica Acta, 237, pp. 79-90 Lee, J.M., Bernstein, A., "Apoptosis, cancer and the p53 tumour suppressor gene" (1995) Cancer and Methastasis Reviews, 14, pp. 149-161 Fung, C.Y., Fisher, D.E., "p53: From molecular mechanisms to prognosis in cancer" (1995) Journal of Clinical Oncology, 13, pp. 808-811 Lane, D.P., Crawford, L.V., "T antigen is bound to a host protein in SV40-transformed cells" (1979) Nature, 278, p. 261 Prokocimer, M., Rotter, V., "Structure and function of p53 in normal cells and their aberrations in cancer cells: Projection on the hematologic cell lineages" (1994) Blood, 84, pp. 2391-2411 Kuerbitz, S.J., Plunkett, B.S., Walsh, W.V., Kastan, M.B., "Wild-type p53 is a cell cycle checkpoint determinant following irradiation" (1992) Proceedings of the National Academy of Sciences of the USA, 89, pp. 7491-7495 Kastan, M.B., Onyekwere, O., Sidransky, D., Vogelstein, B., Craig, R.W., "Participation of p53 protein in the cellular response to DNA damage" (1991) Cancer Research, 51, pp. 6304-6311 Hollstein, M., Sidransky, D., Vogelstein, B., Harris, C.C., "p53 mutations in human cancers" (1991) Science, 253, pp. 49-53 Bosari, S., Viale, G., "The clinical significance of p53 aberrations in human tumours" (1995) Virchows Archives, 427, pp. 229-241 Velculescu, V.E., El-Deiry, W.S., "Biological and clinical importance of the p53 tumor suppressor gene" (1996) Clinical Chemistry, 42, pp. 858-868 Imamura, J., Miyoshi, I., Koeffler, P., "p53 in hematologic malignancies" (1994) Blood, 84, pp. 2412-2421 Guinn, B., Padua, R.A., "Do p53 mutations have a role in the initiation and progression of leukemia?"
