dc.descriptionThe purpose of this study was to investigate the presence of Histoplasma capsulatum in the Kaklik Cave, Denizli, Turkey, newly opened to tourism. The cave has unusual biochemical indigrients causing Pamukkale (Hierapolis)-like travertains. Bat droppings picked from deep parts of the cave were cultured on yeast extract-phosphate agar. Exoantigen and yeast converting tests of suspect colonies containing histoplasma-like macroconidia were performed. These tests were negative. Although DNA encoding partial portion of the M-protein was amplified by PCR, suggesting the isolate to be H. capsulatum, universal primers based on highly conserved regions of the rDNA were used for PCR amplification of the ITS1-5.8S-ITS2 rDNA region followed by sequence comparison to determine phylogenetic relationship of this isolate with fungal species. The sequence data showed 98% homology of this isolate with Renispora flavissima. These results show the importance of traditional characterization by analysis of phenotypes with molecular technology for identification of fungi.
