dc.descriptionC. elegans is an animal model widely used to investigate how neural circuits and genes generate behaviour in response to sensory cues. Inatraditional nose touch assay, C. elegans avoids mechanical stimuli delivered to the nose by moving backwards. Although traditional ways have provided a wealth of knowledge about mechanosensitive behaviours and the molecules mediating it, high-resolution imaging of neuronal activity and synaptic transmission during mechanical nociception remains a challenge. In this project, microfluidic devices were fabricated to partially immobilize individual animals for high-resolution imaging of neuronal activity while delivering a mechanical and chemical stimulus to the nose, to investigate the response of a mechanoreceptor neural pathway. Devices consist of a trapping channel, which allows observing neural responses at the same time a stimulus is delivered with a pneumatic actuated elastic membrane. To evaluate the device performance, it was necessary to measure the membrane displacement using different pressures and compared them with analytical predictions and computational modelling. Additional findings were that the potential intersection of pain and food signals inhibits behavioural responses and calcium activity in the animals, and the role of the serotonin as aneuromodulator in these processes.
