dc.description.abstract | El Factor de Necrosis Tumoral alfa es una citoquina pro-inflamatoria que juega un rol central en la respuesta inmune innata y adaptativa de los primates. Los niveles de expresión del gen se encuentran estrechamente regulados a través de un promotor que se extiende 1,2Kb río arriba del inicio de la transcripción. Los análisis de genómica comparada entre promotores de 5 especies de Platirrinos y 25 especies de Catarrinos, han indicado la presencia de una región de 200pb altamente conservadas entre primates. Este phylogenetic footprinting sería clave para la activación del gen; además los autores indicaron que, por fuera de este segmento, numerosos SNPs se acumulan respetando un modelo de evolución neutral; de las 332 diferencias o sustituciones fijas encontradas respecto del consenso humano, 41 podrían tener valor taxonómico por ser especie-específicas. Se caracterizó la secuencia del promotor del TNF-α en primates del género Sapajus de Argentina: Sapajus nigritus y S. cay. Este trabajo permitió generar el primer reporte de TNF-α en primates neotropicales de la Argentina. Estos datos podrían ser útiles en estudios de genética comparada y evolución del sistema inmune. | |
dc.description.abstract | Tumor Necrosis Factor alpha is a proinflammatory cytokine that plays a central role in the innate and adaptive immune responses in primates. The gene expression levels are tightly regulated by a promoter extending 1.2 kb upstream from the transcription start. The comparative genomic analysis of promoters between 5 Platyrrhine and 25 species of Catarrhini species (including Homo sapiens) have indicated the presence of a 200bp region highly conserved between primates. This phylogenetic footprinting would be key to gene activation. Authors also indicated that variation in genomic regions neighboring this segment, accumulate a number of SNPs under the neutral model of evolution, where 41 SNP, out of 332 substitutions compare with the human genome referes, could be valuable taxonomic being species-specific. The objective of this work is to characterize the promoter sequence of Tumor Necrosis Factor alpha in primates of Sapajus genus in Argentina: Sapajus nigritus (Cebus apella nigritus) and S. cay (Cebus apella Cebus paraguayanus = libidinosus). This study generates the first report of TNF-α neotropical primate of Argentina. These data may be useful in genetic studies and comparative evolution of the immune system. | |
