| dc.description | La Proteína 15 Morfogénica Ósea (BMP15), también conocida como Factor 9B de Crecimiento y Diferenciación (GDF9B), es miembro de la superfamilia β de factores de crecimiento (TGFβ) y su expresión en el ovocito es esencial para el desarrollo y crecimiento folicular. Diferentes polimorfismos en el gen BMP15 han sido asociados con el incremento de tasa ovulatoria y/o prolificidad en algunas razas de ovinos. De esta manera, los objetivos para este estudio fueron determinar tres polimorfismos (FecXG, FecXL y FecXH) con una mutación de una sola base (SNPs) en el gen BMP15, y estimar la asociación de los tres polimorfismos con el tipo de parto (prolificidad) en ovejas de la raza Pelibuey. Se tomaron muestras sanguíneas de la vena yugular de 253 ovejas en edad reproductiva, de acuerdo a la aptitud de producción (MPPA de prolificidad). Mediante la técnica tetraprimer ARMS-PCR (tetra primer amplification refractory mutation system) se determinaron, por primera vez, los tres polimorfismos en las ovejas Pelibuey, y en asociación con los 1050 partos, los genotipos homocigoto silvestre (CC) y mutado (TT) del polimorfismo FecXG con un 45 % y 43 %, respectivamente; y el homocigoto silvestre (GG) y mutado (AA) del polimorfismo FecXL con un 66 % y 29 %, respectivamente; se asociaron a un mayor número de partos dobles (P<0.01). Para el polimorfismo FecXH, solo se encontró el genotipo homocigoto silvestre (CC) con un 51.3 % de parto doble (P<0.01). Los exones 1 y 2 del gen BMP15 fueron amplificados mediante la técnica de Polimerase Chain Reaction (PCR) y secuenciados para la búsqueda de las mutaciones en los polimorfismos (FecXG, FecXL y FecXH), confirmando su ubicación en el exón 2. _______________ POLYMORPHISMS OF THE BONE MORPHOGENETIC PROTEIN 15 (BMP15) ASSOCIATED TO LAMBING TYPE IN THE PELIBUEY EWE. Different BMP15 gene polymorphisms have been associated with increase ovulation rate and/or prolificacy in some sheep breeds. Thus, the aims of this study were to determine three polymorphisms (FecXG, FecXL y FecXH) with a single base mutation (SNPs) in the BMP15 gene, and to estimate the association of the three polymorphisms with lambing type in ewes of the Pelibuey breed. Blood samples were taken by jugular venipuncture from 253 sheep in reproductive age, according to the Most Probable Producing Ability (MPPA of prolificacy). By tetraprimer ARMS-PCR technique (tetra amplification refractory mutation system first), three polymorphisms were determined for the first time in the Pelibuey ewes, and in association with 1050 deliveries, wild homozygous genotype (CC) and mutated (TT) FecXG polymorphism with a 45 % and 43 %, respectively, were associated with a greater number of twining lambing (P <0.01). For FecXH polymorphism, it was only found genotype homozygous wild FecXH (CC) with 51.3% twining lambing (P <0.01). Exons 1 and 2 of the BMP15 gene were amplified by Polimerase Chain Reaction (PCR) technique and sequenced to search of mutations in the polymorphisms (FecXG, FecXL and FecXH), confirming its location in exon 2. | |
