Potencial leishmanicida e citotoxicidade de 4-fenil-1-[2- (ftalimido-2-il)etil]-1h-1,2,3-triazol

As leishmanioses são doenças prevalentes negligenciadas que afetam à saúde pública de forma preocupante e estão cada vez mais presentes nos países em desenvolvimento. Dentre o arsenal químico e farmacológico que emerge na busca de novos compostos leishmanicidas, o derivado ftalimido-triazólico, fenil-1-[2-(ftalimido-2-il)etil]-1h-1,2,3-triazol, foi identificado como composto promissor para obtenção de um novo fármaco contra a leishmaniose. O presente estudo, assim, investigou o efeito in vitro do fenil-1-[2-(ftalimido-2-il)etil]-1h-1,2,3-triazol sobre duas formas evolutivas de Leishmania amazonensis e Leishmania braziliensis. Foi avaliada a citotoxicidade contra as cepas de L. amazonensis, L. braziliensis, e macrófagos peritoneais, bem como a produção de óxido nítrico (NO). Foram realizadas atividades contra amastigotas e formas promastigotas de L. amazonensis e L. braziliensis e investigadas as alterações ultraestruturais nos parasitas e alvos intracelulares envolvidos. Os resultados mostram que o composto obtido da hibridação de ftalimida e 1,2,3-triazol apresentou atividade leishmanicida promissora. O composto fenil-1-[2-(ftalimido-2-il)etil]-1h-1,2,3-triazol produziu alterações ultraestruturais, como encolhimento do corpo celular, perda de integridade da membrana e alterações significativas na morfologia celular e do flagelo . Os resultados mostraram que o composto diminuiu a sobrevivência de amastigotas e apresentou baixa toxicidade para as células hospedeiras. Não houve a indução do aumento de NO nas células tratadas com o composto. De acordo esta pesquisa, o híbrido fenil-1-[2-(ftalimido-2-il)etil]-1h-1,2,3-triazol apresenta efeito leishmanicida e se constitui de um protótipo para o desenvolvimento de novos fármacos anti-leishmania. CNPq Leishmaniasis is a prevalent neglected disease that affects public health in a worrying way and is increasingly present in developing countries. Among the chemical and pharmacological arsenal emerging in the search for new leishmanicidal compounds, the phthalimido-triazole derivative, phenyl-1- [2- (phthalimido-2-yl) ethyl] -1H-1,2,3-triazole, has been identified as a promising compound for obtaining a new drug against leishmaniasis. The present study thus investigated the in vitro effect of phenyl-1- [2- (phthalimido-2-yl) ethyl] -1H-1,2,3-triazole on two evolutionary forms of Leishmania amazonensis and Leishmania braziliensis. The cytotoxicity against strains of L. amazonensis, L. braziliensis and peritoneal macrophages, as well as the production of nitric oxide (NO) were evaluated. Activities were performed against amastigotes and promastigotes of L. amazonensis and L. braziliensis and investigated the ultrastructural changes in parasites and intracellular targets involved. The results show that the compound obtained from the hybridization of phthalimide and 1,2,3-triazole showed promising leishmanicidal activity. The phenyl-1- [2- (phthalimido-2-yl) ethyl] -1H-1,2,3-triazole compound produced ultrastructural changes such as shrinkage of the cell body, loss of membrane integrity and significant changes in cell morphology and of the flagellum. The results showed that the compound decreased the survival of amastigotes and showed low toxicity to the host cells. There was no induction of NO increase in infected cells treated with the compound. According to this research, the phenyl-1- [2- (phthalimido-2-yl) ethyl] -1H-1,2,3-triazole hybrid has leishmanicidal effect and is a prototype for the development of new anti- leishmania drug.
