Purificação, caracterização e aplicação biotecnológica de proteína de folha de Indigofera suffruticosa

Indigofera suffruticosa é uma leguminosa encontrada na região do Agreste Pernambucano, sendo utilizada popularmente como antiespasmódico, diurético e sedativo. O objetivo deste estudo foi isolar, caracterizar estruturalmente e avaliar as aplicações biotecnológicas de uma proteína de folhas de I. suffruticosa. A proteína foi purificada em um único passo por cromatografia de afinidade e apresentou uma massa molecular aparente de 32 KDa e pI de 7.09 e 7.94. Essa proteína apresentou maior atividade hemaglutinante específica (14,629) para eritrócitos humanos do tipo A e foi inibida por glicoproteínas e monossacarídeos, demonstrando ser uma lectina. Sua atividade hemaglutinante não foi alterada por cátions divalentes, foi estável na faixa de pH entre 6,5 a 7,0 e termoresistente. Espectroscopia de fluorescência intrínseca e extrínseca demonstrou a presença de resíduos de triptófano e tirosina expostos na superfície da molécula e regiões hidrofóbicas parcialmente internalizadas. Espectrometria de massa revelou similaridade com uma proteína de Panicum hallii. A espectroscopia de infravermelho revelou que a lectina possui um maior percentual de estruturas do tipo folhas β. Desconvolução do espectro de dicroísmo circular indicou a presença de 23% de α-hélices, 30% de folhas-β, 20% de voltas- β e 27% de estruturas desordenadas, pertercendo à classe de estrutura terciária α+β. A proteína mostrou potencial antioxidante máximo de 52 % (1mg/mL) no ensaio de redução do radical, 2,2-difenil-1-picril-hidrazil (DPPH). No ensaio do sequestro do radical 2,2-azino-bis (3-etilbenzotiazolina-6 ácido sulfônico) (ABTS) a proteína apresentou porcentagem de inibição de 36,71% após 120 min., o que corresponde a um CAET de 0,698 μmol/ mg. A proteína mostrou poder redutor, indicando transferência de elétrons como o principal mecanismo antioxidante. A lectina demonstrou ação bacteriostática contra Staphylococcus aureus (CIM = 15 μg mL⁻¹) e fungistática contra Candida krusei, Candida tropicalis, Candida parapsilosis e Candida albicans com CIM de 8, 16, 32 e 64 μg mL⁻¹, respectivamente. A proteína apresentou baixa atividade hemolítica com porcentagem de hemólise variando entre 0,1% a 20,4% nas concentrações de 0,062 a 4 mg/mL. A lectina mostrou- se como agente que prolonga a via intrínseca da coagulação sanguínea. Os resultados indicam que a proteína isolada de folhas de I. suffruticosa, é uma lectina de estrutura estável, com atividade antioxidante, antimicrobiana e anticoagulante, de potencial aplicação biotecnológica. FACEPE Indigofera suffruticosa is a legume found in the region of the Agreste Pernambucano, being popularly used as antispasmodic, diuretic and sedative. The objective of this study was to isolate, characterize structurally and evaluate the biotechnological applications of a protein of leaves of I. suffruticosa. The protein was purified in a single step by affinity chromatography and showed an apparent molecular mass of 32 KDa and pI of 7.09 and 7.94. This protein showed higher hemagglutinating activity specific (14.629) for human type A erythrocytes and was inhibited by glycoproteins and monosaccharides, proving to be a lectin. Its hemagglutinating activity was not altered by divalent cations, was stable in the pH range between 6.5 and 7.0 and thermoresistant. Intrinsic and extrinsic fluorescence spectroscopy demonstrated the presence of exposed tryptophan and tyrosine residues on the surface of the molecule and partially internalized hydrophobic regions. Mass spectrometry revealed similarities with a Panicum hallii protein. Infrared spectroscopy revealed a higher percent of β-sheet structures in the lectin molecule. Deviation of the circular dichroism spectrum indicated the presence of 23% α-helices, 30% β-sheets, 20% β-turns and 27% of disordered structures, belonging to the α + β tertiary structure class. The protein showed a maximum antioxidant potential of 52% (1mg / mL) in the radical reduction assay, 2,2-diphenyl-1-picrylhydrazyl (DPPH). In the assay of sequestration of the 2,2-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical the protein showed a 36.71% inhibition percentage after 120 minutes, corresponding to a CAET of 0.698 μmol/mg. The protein showed reducing power, indicating electron transfer as the main antioxidant mechanism. Lectin demonstrated bacteriostatic action against Staphylococcus aureus (MIC = 15 μg mL⁻¹) and fungistatic against Candida krusei, Candida tropicalis, Candida parapsilosis and Candida albicans with MIC from 8, 16, 32 and 64 μg mL⁻¹, respectively. The protein showed low hemolytic activity with hemolysis percentage ranging from 0.1% to 20.4% at concentrations of 0.062 to 4 mg/mL. Lectins showed as an agent that prolongs the intrinsic pathway of blood coagulation. The results indicate that the protein isolated from leaves of I. suffruticosa is a stable structure lectin with antioxidant, antimicrobial and anticoagulant activity of potential biotechnological application.
