dc.description.abstractA semi-nested PCR was standardized to amplify the gG coding region of Bovine Herpesvirus (BpHV). This PCR was able to identify standard sample of BoHV-1.1 (Colorado ATCC 864), BoHV-1.2 (K22) and BoHV-5 (EVI-88). This test was also efficient in indentifying infection of cell cultures inoculated with material from semen, nasal swabs and central nervous system (CNS). The efficiency of the test was evaluated by restriction endonuclease assay with XhoI and by the amplicons sequencing. The genic region used showed to be efficient in identifying BoHV-1 and BoHV-5 in samples. For the regions and conditions used, the Heteroduplex Mobility Assay (HMA) showed to be unsatisfactory for differentiating subgroups. A co-infection of Bovine Syncytial Virus (BRSV) and BoHV-5 was detected in cows of a dairy farm. This report has alsoshowed BoHV-5 envolved in respiratory disorder in adult animals
