Growing and finishing pigs are affected by pathogenic spirochetes of the genus Brachyspira sp., which cause a significant economic impact due to direct and indirect losses. Thus, efficient diagnosis of these species enables better technical intervention to prevent or treat diseases. This study aimed to evaluate the efficiency of isolation, PCR, FISH and IHC for the diagnosis of B. hyodysenteriae and B. pilosicoli. Thirty -nine samples were collected from pigs positive for this species in at least one test. The number of positives in each assay was evaluated in a comparative form, using the McNemar statistical method, with p<0.05. Out of the 39 analyzed samples, three (7,69%) were positive by isolation, 28 (71.7%) by PCR and FISH and 32 (82.0%) by IHC. The bacterial isolation had the lowest rate of detection, statistically inferior to the other tests. Based on the practicity, speed and association with histologic lesions, FISH has become a di agnostic alternative for Brachyspira sp..
