dc.description.abstract | La porfiria se refiere a desordenes genéticos que afectan la biosíntesis del grupo heme causando acumulación de precursores de la ruta de síntesis de las porfirinas. La Porfiria Aguda Intermitente es una enfermedad autosómica dominante con penetrancia incompleta, que resulta de una deficiencia en la enzima porfobilinógeno deaminasa (PBGD). El gen HMBS contiene la información para la síntesis de la enzima PBGD y un déficit de esta causa acumulación de ácido delta-aminolevulínico y porfobilinógeno en el hígado. En este gen se han encontrado más de 300 mutaciones que llevan al fenotipo de porfiria aguda intermitente. Este proyecto busca encontrar la relación entre el fenotipo y el genotipo de una familia colombiana diagnosticada previamente con porfiria intermitente aguda a lo largo de sus generaciones. Se usaron muestras de sangre de la familia para la extracción de ADN con el cual se realizó la amplificación de las muestras por PCR de los exones 11, 12, 13, 14 y 15 del gen HMBS. A partir de esto se identificó la posible variante patogénica situada en la región conservada de fin del splicing antes del exón 15 en el individuo 1 y 3 de la familia | |
dc.description.abstract | Porphyria refers to genetic disorders that affect the heme biosynthesis causing accumulation of precursors of the porphyrin synthesis pathway. Acute Intermittent Porphyria is an autosomal dominant disease with incomplete penetrance, which results from a deficiency in the porphobilinogen deaminase enzyme (PBGD). The HMBS gene contains the information for the synthesis of the enzyme PBGD and a deficit of this cause accumulation of delta-aminolevulinic acid and porphobilinogen in the liver. In this gene, more than 300 mutations have been found that lead to the phenotype of acute intermittent porphyria. This project seeks to find the relationship between the phenotype and the genotype of a Colombian family previously diagnosed with acute intermittent porphyria throughout their generations. Blood samples from the family were used for the extraction of DNA with which the amplification of the samples was carried out by PCR of exons 11, 12, 13, 14 and 15 of the HMBS gene.
