Las infecciones por hantavirus en humanos causan enfermedades zoonóticas que se presentan al estar en contacto con fluidos corporales y/o con heces secas de ratones infectados. La familia de los Hantaviridae está presente en todo el mundo y, aunque no todos los hantavirus causan enfermedad en humanos, de aquellos que sí las causan se ha reportado en estudios previamente realizados que las proteínas NS producidas por estos están involucradas en la inhibición del factor regulador de interferón 3 (IRF3), precursor de Interferón B. Partiendo de esto, en este proyecto se seleccionaron 5 secuencias de las proteínas NS, 2 de hantavirus patógenos, 2 de hantavirus no patógenos y 1 del virus colombiano Necocli orthohantavirus del cual no se ha determinado patogenicidad. Esta selección se dio para evaluar si hay o no diferencias en el comportamiento de los virus patógenos y no patógenos, además de observar el comportamiento del virus colombiano. A partir de estas secuencias, se utilizó el software I-Tasser para realizar el plegamiento por homología de estas proteínas, mientras que la estructura de la forma libre de la proteína IRF3 se seleccionó de Protein Data Bank. Posteriormente, para cada proteína, se realizaron dinámicas moleculares y con los resultados de estas se llevó a cabo el docking de cada una de las proteínas NS con el IRF3. Estos resultados mostraron una diferencia entre el tipo de interacción presentado ya que, las proteínas de los virus patógenos interactuaban en la región del pocket de la proteína precursora, mientras que las no patógenas interactuaban fuera de este. Para la proteína colombiana se obtuvo una interacción con la parte externa de este. Con los resultados se logró establecer que hay una diferencia entre la interacción de las proteínas NS, patógenas y no patógenas, y la proteína IRF3. Adicionalmente, al evaluar el comportamiento de la proteína NS del virus colombiano, se obtuvo que es similar al de las proteínas patógenas evaluadas en este estudio. The Hantaviridae family is present around the world and, even when not all they cause disease in humans, from those that are pathogens, is not known the specific mechanism by which they evade the immune system, even though previous studies have shown that the NS proteins produced by hantaviruses are involved in the inhibition of Interferon Regulatory Factor 3(IRF3), a precursor of Interferon B. Given what has been said, we chose 5 protein sequences of the hantavirus protein NS, 2 from pathogenic viruses, 2 from not pathogenic viruses and 1 from a Colombian virus Necocli orthohantavirus, whose pathogenicity has not been determined. This selection was made to evaluate whether there are differences in the behavior of pathogenic and non-pathogenic viruses, in addition to observing the behavior of the Colombian virus. Following this, using the software I-Tasser we performed the homology folding of these 5 proteins, while the structure of the free form of the IRF3 was selected from Protein Data Bank. Subsequently, for each protein, we did molecular dynamics and with the results of this procedure, we made a docking process between the NS proteins and the IRF3. These results showed a difference between the type of interaction presented among the NS proteins and the precursor, where the pathogenic viruses proteins interacted in the pocket region of the precursor protein, while the non-pathogenic ones interacted outside of it, for the Colombian protein, we had obtained an interaction with IRF3's external part.
