Orientador: Marcelo Brocchi Tese (doutorado) - Universidade Estadual de Campinas, Instituto de Biologia Resumo: Salmonella enterica é o principal agente etiológico causador de doenças de origem alimentar em humanos. Dentre os mais de 2600 sorovares de Salmonella, destaca-se Salmonella enterica subespécie enterica sorovar Enteritidis, devido ao aumento significativo de surtos causados por este sorovar nos últimos anos em vários países, inclusive no Brasil. Em virtude da importância deste patógeno na saúde pública, uma das formas que vem sendo estudada para o controle de infecções é o uso de vacinas. Neste trabalho, construímos e caracterizamos mutantes para genes codificadores de proteínas associadas ao nucleóide (Naps) de Salmonella enterica sorovar Enteritidis PT4. As características fenotípicas dos mutantes nulos de Salmonella Enteritidis para os genes ihfA ou ihfB, codificadores das subunidades 'alfa' e ß de IHF, foram avaliadas quanto ao crescimento, motilidade, sobrevivência em estresse oxidativo e quanto à capacidade de invasão e sobrevivência em macrófagos. Testes de atenuação e avaliação da capacidade de induzir proteção em caso de infecção por S. enterica foram realizados utilizando o modelo murino. Análises da expressão dos genes ihfA e ihfB também foram realizadas na linhagem selvagem e nos mutantes ihf. Nossos dados revelam que mutantes ihf mostraram uma pequena redução na taxa de crescimento in vitro e uma diminuição significativa na motilidade. Além disso, o mutante 'delta'ihfA tornou-se sensível a presença de H2O2 e apresentou uma redução na capacidade de invadir e sobreviver no interior de macrófagos, enquanto que o duplo mutante tornou-se mais resistente a presença de H2O2 e teve sua capacidade de invadir e sobreviver no interior de macrófagos aumentada quando comparado com a linhagem selvagem. Os mutantes de S. enterica Enteritidis para os genes ihfA e ihfB foram atenuados e, portanto, passíveis de serem testados como vacinas vivas atenuadas. Tais mutantes atenuados foram capazes de induzir proteção em modelo murino, quando desafiadas com a linhagem selvagem. Entretanto, o duplo mutante não foi capaz de induzir proteção. Além disso, ambos os genes foram mais expressos na transição para a fase estacionária na linhagem selvagem. Em mutantes 'delta'ihfB, o gene ihfA foi super-expresso e a presença do sitio de ligação da proteína IHF na região promotora do gene ihfA, revela uma auto regulação negativa por IHF. Esses resultados indicam que IHF atua como um repressor para ihfA e possui um papel importante na regulação positiva do crescimento, motilidade e patogenicidade em Salmonella Enteritidis. Além disso, mostram claramente que os mutantes nulos simples aqui desenvolvidos têm potencial para serem avaliados como vacinas vivas atenuadas Abstract: Salmonella enterica is one of the major agents of intestinal infections worldwide. This species is classified in more than 2600 serovars, been S. enterica Enteritidis the more frequently serovar involved with infections in humans and other animals. This infection can progress to serious extra-intestinal infections which bring the control of salmonelosis a priority. One of the means to control these infections is the development of vaccines. Based on this, the aim of this work was to construct strains of S. enterica Enteritidis PT4 mutants for Naps (Nucleoid Associated Proteins). Phenotypic characteristics of the ihfA or ihfB mutants, genes encoding the ? and ? subunits of Integration Host Factor (IHF) such as growth in vitro, motility, oxidative stress resistance, invasion and multiplication in macrophages were evaluated. In addition, virulence and the ability to induce protection in the mouse model of infection were assayed. The ihfA and ihfB expressions were also assayed. The mutants presented a light reduction in the in vitro growth but a significant reduction in motility. The S. enterica Enteritidis PT4 'delta'ihfA strain was more sensitive to hydrogen peroxide and exhibited a reduced capacity to invade and to survive into macrophages in culture. On the contrary, the 'delta'ihfAB strain exhibited resistance to hydrogen peroxide and a superior capacity to invade and survive intracellularly when compared to the wild type isogenic strain (WT). The three mutant strains ('delta'ihfA, 'delta'ihfB and 'delta'ihfAB) were attenuated in the mouse model of infection but only the single mutants were able to induce protection after challenge with a lethal dose of the WT strain. Transcription of ihf genes was observed in the transition of logarithmic to stationary phase. In ihfB mutant strain, ihfA gene transcription was induced indicating that IHF inhibits ihfA expression. Our data suggest that IHF has an important role in the biology of S. enterica Enteritidis.
