PRODUCTION AND CHARACTERIZATION OF EXTRACELLULAR CELLULASES FROM A THERMOSTABLE ASPERGILLUS SP

Thermostable Aspergillus sp., 10-2, was isolated from compost heap and the temperature range for growth was 20-50-degrees-C and the optimum growth temperature range was 35-45-degrees-C. Highest production of endo-1,4-beta-D-glucanase EC. 3.2.1.4 (CMCase), exo-1,4-beta-D-cellobiohydrolase EC. 3.2.1.91 (avicelase), and beta-glucosidase EC. 3.2.1.21 (cellobiase) was obtained when the mold was grown on culture medium containing paper cellulose powder. The strain produced more CMCase at 37-degrees-C as compared to other temperature and incubation at 45-degrees-C produced remarkably high activity of beta-glucosidase. The optimum temperature for saccharification of cellulose by the enzyme was 55-degrees-C. Paper chromatographic analysis of cellulose hydrolysates demonstrated predominantly glucose as compared to cellulose hydrolysates by Trichoderma reesei QM 9414 enzyme. This is due to fact that the strains of Aspergillus sp., 10-2 produced more beta-glucosidase. 23 3 183 188
