Resumen
The gerbil is a rodent considered a good model for studies of prostatic morphophysiology under different experimental conditions. Studies involving castration and steroidal blockers of aged gerbils showed that the glandular epithelium persists after long-term therapy, preventing the organ atrophy. Thus, the objective of this study was to evaluate the phenotypic characteristics and behavior of prostatic epithelial cells that remained after different periods of hormone ablation in aged gerbils. The identification of elements that influenced the survival of this cell type was performed by morphometric, nuclear phenotypes, ultrastructural and immune histochemical analysis. The most significant responses to treatment, by analyzing morphometric features, were observed during the first three time points (day 1, day 3, and day 7), after which there appeared to be an adjustment of the gland to the hormone ablation. All treatments led to changes in the state of chromatin condensation, DNA methylation pattern and phenotypic changes indicated cell senescence. Additionally, an increase in the basal cells seemed to guarantee self-renewal properties to the epithelium. These data indicate that changes occur at many levels, including gene expression and nuclear architecture in the epithelial cells, when aging and steroidal blockade are associated. These aspects are important when considering castration-resistant prostate cancer, a malignant tumor posing difficult therapeutic intervention. © 2013 Elsevier GmbH. 116 1 204 213 Abate-Shen, C., Shen, M.M., Molecular genetics of prostate cancer (2000) Genes Dev, 14, pp. 2410-2434 Arcolino, F.O., Ribeiro, D.L., Gobbo, M.G., Taboga, S.R., Góes, R.M., Proliferation and apoptotic rates and increased frequency of p63-positive cells in the prostate acinar epithelium of alloxan-induced diabetic rats (2010) Int J Exp Pathol, 91, pp. 144-154 Augusto, T.M., Bruni-Cardoso, A., Damas-Souza, D.M., Zambuzzi, W.F., Kühne, F., Lourenço, L.B., Oestrogen imprinting causes nuclear changes in epithelial cells and overall inhibition of gene transcription and protein synthesis in rat ventral prostate (2010) Int J Androl, 33, pp. 675-685 Banerjee, P.P., Banerjee, S., Brown, T.R., Castration-induced apoptotic cell death in the Brown Norway rat prostate decreases as a function of age (2000) Endocrinology, 141, pp. 821-832 Banerjee, P.P., Banerjee, S., Lai, J.M., Strandberg, J.D., Zirkin, B.R., Brown, T.R., Age-dependent and lobe-specific spontaneous hyperplasia in the Brown Norway rat prostate (1998) Biol Reprod, 59, pp. 1163-1170 Banerjee, P.P., Banerjee, S., Tilly, K.I., Brown, T.R., Zukin, B.R., Lobe-specific apoptotic cell death in rat prostate after androgen ablation by castration (1995) Endocrinology, 136, pp. 4368-4376 Bornman, D.M., Mathew, S., Alsruhe, J., Herman, J.G., Gabrielson, E., Methylation of the E-cadherin gene in bladder neoplasia and in normal urothelial epithelium from elderly individuals (2001) Am J Pathol, 159, pp. 831-835 Bruyninx, M., Hennuy, B., Cornet, A., Houssa, P., Daukandt, M., Reiter, E., A novel gene overexpressed in the prostate of castrated rats: hormonal regulation, relationship to apoptosis and to acquired prostatic cell androgen independence (1999) Endocrinology, 140, pp. 4778-4799 Burzynski, S.R., Aging: gene silencing or gene activation?
