Resumen
Es importante diseñar técnicas eficientes, prácticas y fáciles de
realizar para valorar la integridad acrosomal (VIA) de los espermatozoides
(EPZ). El objetivo fue evaluar la modificación de la
técnica de tinción Giemsa para valoración de VIA en EPZ de
mamíferos. Se evaluaron 140 laminillas con EPZ de 4 diferentes
mamíferos (bovinos, ovinos, porcinos y humanos), se dividieron
en dos grupos (70 laminillas por grupo). El primer grupo, evaluó
VIA con la técnica clásica de tinción Giemsa y el segundo, con
la técnica modificada de Giemsa. Se compararon las medias de
VIA y los tiempos de cada una de las técnicas. No hubo diferencia
entre las medias de VIA en ambas técnicas; la técnica modificada
redujo en 90 minutos el tiempo de realización y mejor claridad de
la imagen. En conclusión, la técnica de tinción modificada disminuye
el tiempo de valoración de VIA y muestra una mejor nitidez
de la imagen. It is important to design efficient, practical, and easy-to-perform
techniques to assess the acrosomal integrity (AAI) of sperm antes
de (SPZ). The objective was to evaluate the modification of the
Giemsa staining technique for AAI assessment in mammalian
SPZ. One hundred fourty SPZ flakes from 4 different mammals
(cattle, sheep, pigs and humans) were evaluated, they were
divided into two groups (70 flakes per group). In the first group, AAI
was evaluated with the classical Giemsa staining technique and
the second with the modified Giemsa technique. The AAI means
and the times of each of the techniques were compared. There
was no difference between the AAI means in both techniques;
the modified technique reduced the execution time and improved
image clarity by 90 minutes. In conclusion, the modified staining
technique decreases the AAI assessment time and shows better
