Relación entre el polimorfismo de k-caseína y B-lactoglobulina con el contenido de calcio, fósforo, citrato y termoestabilidad de la leche

El objetivo del presente trabajo fue determinar la relación entre el polimorfismo de la к-caseína y β-lactoglobulina de la leche y el contenido de calcio, fósforo, citrato y termoestabilidad en leches de 10 vacas Frisón Negro, durante el período de primavera del 2001. Las muestras de leche de vacas Frisón Negro se obtuvieron del fundo Santa Rosa, Provincia de Valdivia, Décima Región de Los Lagos, propiedad de la Universidad Austral de Chile. Para la determinación de las variantes genéticas de к-caseína y β-lactoglobulina se realizó una electroforesis de isoenfoque. La obtención de la к-caseína de la leche, fue mediante la separación de la к-caseína de las caseínas sensible al calcio por calentamiento a 35°C. La obtención de las muestras de β-lactoglobulina se realizó en el suero de la leche por centrifugación. En las muestras de leche de к-caseína solamente se pudo identificar las variantes genéticas A o B mediante electroforesis de isoenfoque. Para las muestras de leche de β-lactoglobulina se identificaron la presencia en conjunto de las variantes A y B, presentándose una mayor expresión de la variante A respecto a la B en las vacas de raza Frisón Negro que presentaron el fenotipo AB. Se obtuvo que existen diferencias estadísticamente significativas (p<0,05) entre las variantes genéticas de к-caseína y el contenido de calcio total, fósforo total, citrato total y termoestabilidad en las leches de vacas Frisón Negro. Para la β-lactoglobulina sólo se encontraron diferencias estadísticamente significativas entre el contenido de citrato total y las variantes genéticas identificadas, siendo la variante AA de β-lactoglobulina la que reflejó un mayor contenido de citrato total respecto al fenotipo AB. Además, se encontraron diferencias estadísticamente significativas (p<0,05) entre el contenido de fósforo total, citrato total y los meses de muestreos. Respecto a la к-caseína la variante B obtuvo mayores valores en los contenidos de calcio total, fósforo total y una mayor termoestabilidad respecto a la variante A. Se obtuvo correlaciones significativas entre la termoestabilidad y el contenido de calcio y fósforo total, no así entre la termoestabilidad y el contenido de citrato total y pH. The main objective of the present work is to determine the relationship between the polymormorphism of the к-casein and β-lactoglobulin and the calcium, phosphorus and citrate contents and thermostability in the milk of 10 Black Frison cows. Sampling was done during spring 2001. The milk samples were obtained from the Universidad Austral Santa Rosa farm, Provincia de Valdivia, Décima Región de los Lagos, Universidad Austral de Chile. To determine the genetic variants in к-casein and β-lactoglobulin, an isoelectric focusing was carried. The obtaining of the milk к-casein was fulfilled the separated of the к-casein of the casein sensible at calcium for heating at 35° C. The samples of β-lactoglobulin was obtained in the serum of the milk for centrifugation. In the milk samples of к-casein it was only possible to identify the variants A or B by isoelectric focusing. The presence of both variants A and B was demonstrated after analysis of the milk samples containing β-lactoglobulin. The variant A showed a bigger expression in comparison to the variant B for the Black Frison cows race, which presented the phenotype AB. The results indicated significant statistical differences (p<0,05) between the genetic variants of к-casein and the total calcium, phosphorus and citrate contents and thermostability in the milk of Black Frison cows. For the β-lactoglobulin, significant statistical differences were found only between the total citrate content and the identified genetic variant, being the AA variant of β-lactoglobulin the one that reflected a bigger content of total citrate regarding the phenotype AB. There were also significant statistical differences (p<0,05) between the total phosphorus and citrate contents and the months of sampling. Regarding the к-casein the variant B obtained bigger values in the total calcium contents, total phosphorus and a bigger thermostability regarding the variant A. Significant correlations were obtained between thermostability and the content of total calcium and phosphorus. However, this was not the case for thermostablity, total citrate content and pH.
