Antiplatelet effect of Aristotelia chilensis (maqui) extracts through in vitro studies.

Chile, like other countries, has high mortality rates due to cardiovascular diseases (CVD; eg, acute myocardial infarction, cerebrovascular disease or peripheral thrombosis). The complex pathophysiological process involved in CVD includes the participation of platelets.
It has been observed that some bioactive compounds present in fruits and vegetables that are consumed regularly can inhibit platelet activation and therefore reduce the risk of CVD.
Aristotelia chilensis (Mol.) Stuntz, also known as maqui, is a berry native to central and southern Chile. Maqui has been used for food, cosmetic and pharmacological purposes, however, up to date there are no reports of studies related to its antiplatelet activity. Preliminary studies carried out at the Thrombosis Research Center (CIT), Faculty of Health Sciences, University of Talca, confirmed that ethanolic extracts obtained from A. chilensis fruits (Luna Nueva, Morena and Perla Negra varieties) show an antiplatelet effect in vitro, using ADP 4 µM as an agonist. With the aim of deepening this line of research, a study of the antiplatelet potential of different clones of this species was carried out, together with the participation of possible molecules responsible for this activity, as well as the possible mechanism on mitochondrial function. The antiplatelet activity of three domesticated maqui clones was investigated: Luna Nueva, Morena and Perla Negra. Platelet aggregation was assessed by turbidimetry. Granule secretion and ROS production were studied by flow cytometry. The most active extracts were characterized by HPLC/UV and HPLC-MS/MS. Additionally, the effect of isoramnetin on mitochondrial function, platelet adhesion and thrombus formation under controlled blood flow conditions in the Badimon Perfusion Chamber was evaluated.
The extracts of leaves and immature fruits of the Luna Nueva and Morena clones significantly inhibited the aggregation and platelet secretion induced by ADP 4 µM and collagen1 µg/mL. Furthermore, these extracts reduced oxidative stress. The chemical characterization allowed to identify of several chemical compounds with antiplatelet potential in this berry. Isoramnetin was one of the compounds that presented a positive correlation between its concentration and the antiplatelet potential of the extracts studied. This flavonol showed antiplatelet activity induced by the agonists, collagen and TRAP-6, with IC50 values of 8.1 ± 2.6 µM and 16.1 ± 11.1 µM, respectively, on the other hand, it inhibited activation and platelet secretion at 20 µM. Also, ROS release increased as a consequence of decreased mitochondrial membrane potential. Isoramnetin also reduced platelet deposition on the thrombus, confirming its antithrombotic effect.
This work is the first evidence of antiplatelet activity evaluated in A. chilensis. Only the ripe fruit is used in the pharmaceutical industry, so the evaluation of the leaves and immature fruits gives added value to this species, thus increasing the interest in native plants with biological properties. Chile, como otros países, tiene elevadas tasas de mortalidad por enfermedades cardiovasculares (ECV; ej. Infarto agudo de miocardio, enfermedad cerebrovascular y trombosis periféricas). El complejo proceso fisiopatológico involucrado en las ECV incluye la participación de las plaquetas.
Se ha observado que algunos compuestos bioactivos presentes en frutas y verduras que se consumen regularmente pueden inhibir la activación plaquetaria y por tanto reducir el riesgo de ECV.
Aristotelia chilensis (Mol.) Stuntz, también conocido como maqui, es una baya que crece en el centro y sur de Chile. El maqui ha sido usado con fines alimenticios, cosméticos y farmacológicos, sin embargo, hasta la fecha no hay informes de estudios relacionados con la actividad antiplaquetaria. Estudios preliminares realizados en el Centro de Investigaciones en Trombosis (CIT), Facultad de Ciencias de la Salud, Universidad de Talca, confirmaron que extractos etanólicos obtenidos a partir de frutos de A. chilensis (variedades Luna Nueva, Morena y Perla Negra) presentan efecto antiplaquetario in vitro, usando ADP 4 µM como agonista. Con el objetivo de profundizar en dicha línea de investigación se realizó un estudio del potencial antiplaquetario de diferentes clones de esta especie, además se investigó la participación de posibles moléculas responsables de esta actividad, así como el posible mecanismo sobre la función mitocondrial. Se investigó la actividad antiplaquetaria de tres clones de maqui domesticadas: Luna Nueva, Morena y Perla Negra. Se evaluó la agregación plaquetaria mediante turbidimetría. La secreción de gránulos y producción de ROS se estudiaron por citometría de flujo. Los extractos más activos fueron caracterizados por HPLC/UV y HPLC-MS/MS. Adicionalmente, se evaluó el efecto de la isoramnetina sobre la función mitocondrial, adhesión plaquetaria y formación de trombos bajo condiciones de flujo sanguíneo controlado en la Cámara de Perfusión Badimon.
Los extractos de hojas y frutos inmaduros de los clones Luna Nueva y Morena inhibieron significativamente la agregación y secreción plaquetaria inducida por ADP 4 µM y colágeno 1 µg/mL. Además, estos extractos redujeron el estrés oxidativo. La caracterización química permitió identificar varios compuestos químicos con potencial antiplaquetario en esta baya. La isoramnetina fue uno de los compuestos que presentó una correlación positiva entre su concentración y el potencial antiplaquetario de los extractos estudiados. Este flavonol mostró actividad antiplaquetaria inducida por los agonistas, colágeno y TRAP-6, con valores de IC50 de 8,1 ± 2,6, µM y 16,1 ± 11,1 µM, respectivamente, por otro lado, inhibió la activación y secreción plaquetaria a 20 µM. También aumentó la liberación de ROS como consecuencia de la disminución del potencial de membrana mitocondrial. La isoramnetina, además, redujo la deposición de plaquetas sobre el trombo, confirmando su efecto antitrombótico.
