La familia Dual-specificity Tyrosine Regulated Kinases (DYRK) posee actividad
Ser/Thr quinasa y está conformada por cinco miembros clasificados en dos
grupos, DYRK1A y DYRK1B componen la clase l mientras que DYRK2, DYRK3 y
DYRK4 constituyen la clase ll. Estas quinasas poseen similitud en secuencia y
función, se distribuyen en diversos tejidos y presentan diferencias en la
especificidad de sustratos. Las DYRK tienen una alta expresión en testículo, pero
poco se sabe del rol que tendría en dicho órgano. El objetivo de este trabajo fue
analizar el rol de la familia de las DYRK en la regulación por fosforilación del
residuo Ser640 de glucógeno sintasa muscular (MGS), enzima altamente regulada
y que se expresa en células componentes del epitelio seminífero. Además,
evaluamos los efectos de la inhibición farmacológica sobre la viabilidad y motilidad
de espermatozoides. Primero, detectamos la expresión diferencial de las cinco
DYRKs mediante qRT-PCR, observando la alta expresión de DYRK1A en las
líneas celulares Sertoli adulta 42GPA9 y en germinales GC-1 y GC-2 mientras que
a nivel proteico, DYRK1A y DYRK1B son las más abundantes de acuerdo al
análisis por Western Blot. Por inmunofluorescencia y fraccionamiento subcelular
evaluamos la distribución de las DYRK1A, DYRK1B, MGS y pGS (Ser640),
demostrando que estas proteínas comparten el mismo compartimento subcelular y
podrían interactuar de acuerdo al análisis de coinmunoprecipitación. Realizamos la
inhibición farmacológica de DYRK1A y DYRK1B con INDY (Inhibitor of DYRK) así
como también el silenciamiento de ambas enzimas con shRNAs y evaluamos el
estado de fosforilación de Ser640 y el contenido de glucógeno, demostrando que
hay diferencias en la respuesta celular en cada una de las líneas. Con respecto al
efecto de INDY sobre los espermatozoides, vimos que no altera la viabilidad
celular y es capaz de incrementar la progresividad del movimiento. Estos
hallazgos sugieren que DYRK1A y DYRK1B ejercen un rol regulatorio en la
síntesis de glucógeno en Sertoli y células germinales y que ambas quinasas
podrían estar modulando el movimiento del espermatozoide sin afectar su
supervivencia. Dual-specificity Tyrosine Regulated Kinases (DYRKs) family has Ser/Thr kinase
activity and contains five members classified in two groups, DYRK1A and DYRK1B
constitute class l meanwhile, DYRK2, DYRK3 and DYRK4 constitute class ll. They
have sequence and functional similarities, are distributed in many tissues and
present different substrate specificities. DYRKs are highly expressed in testis but
little is known about their roles. The aim of this work was to analyze the role of
DYRK family on the regulation by phosphorylation of Ser640 residue of Muscle
Glycogen Synthase (MGS), enzyme that is highly regulated and expressed in
cellular components of seminiferous epithelium. Additionally, we evaluated the
effects of pharmacological inhibition on sperm viability and motility. First, we
detected differential expression of the five DYRKs by qRT-PCR observing that
DYRK1A is highly expressed in adult Sertoli cell line 42GPA9 and germ cells GC-1,
GC-2. Meanwhile at protein level, DYRK1A and DYRK1B are the most abundant
according to Western blot analysis. Through immunofluorescence and subcellular
fractionation, we evaluated the distribution of DYRK1A, DYRK1B, MGS and pGS
(Ser640) demonstrating that these proteins share the same location in the cell and
could also interact according to coimmunoprecipitation analysis. We performed the
pharmacological inhibition of DYRK1A and DYRK1B with INDY (Inhibitor of DYRK)
as well as gene silencing of both enzymes by shRNAs, and evaluated the
phosphorylation status of Ser640 and glycogen content, showing that there are
differences in the cellular response in each cell line. With regard to the effect of
INDY on sperm, we observed that it does not alter cell viability and is capable to
increase the progressivity of movement. These findings suggest that DYRK1A and
DYRK1B exert a regulatory role in the synthesis of glycogen in Sertoli and germ
cells and that both kinases could be modulating the movement of the sperm
