Mitigación de la patogenicidad del virus tobacco mosaic virus (tmv), en el desarrollo de la planta de tomate (Solanum lycopersicum L.), utilizando nanotecnología

Resumen
"El TMV causa daños graves y pérdidas en la cosecha del 11 al 80%,
particularmente bajo invernadero, donde se calculan pérdidas medias anuales
de hasta un millón de dólares. El objetivo del estudio fue evaluar la
potencialidad bipesticida de 360°Agrocker® contra la patogénesis causada por
TMV en diversas etapas fenológicas en el cultivo de tomate. La investigación se
realizó en la Universidad Autónoma Agraria Antonio Narro, comenzando
primeramente por evaluar la calidad fitosanitaria de semilla de dos genotipos de
tomate a utilizarse en la investigación, VILLA NARRO® y SofiMely®. Estos se
analizaron microbiológicamente para detectar fitopatógenos en semilla, en
medios generales como papa-dextrosa-agar (PDA) y malta sal agar (MSA). Se
identificaron géneros de bacterias fitopatógenas utilizando tinción de Gram,
corroborando con la prueba de Ryu y medios semi-selectivos como CNS y
Levadura-Dextrosa-Calcio (YDC). Se aplicó la prueba serológica de ELISA
(AGDIA®), para detectar TMV en semillas. Se aplicó un análisis estadístico para
hongos con un diseño completamente al azar con arreglo factorial. Se
encontraron diferencias significativas entre el número de hongos detectados
(P=0.05*), incluyendo géneros como: Fusarium spp, Alternaria spp. Penicillium
spp. Aspergillus spp y Cladosporium spp., cuya incidencia fue de 15.83 7.5,
3.33, 4.16 y 1.66 % respectivamente. No se encontraron diferencias
significativas entre los genotipos (P=0.91) y medios de cultivo (P=0.07). En PDA
se desarrollaron cepas bacterianas potencialmente fitopatógenas amarillas y en
MSA anaranjadas, que se asociaron a los géneros Xanthomonas (0.66%) y
Clavibacter (0.33%) respectivamente; la incidencia de bacterias fue baja
(0.74%). Las lecturas de absorbancia del espectrofotómetro, en las muestras de
semilla de la prueba ELISA, resultaron menores que las de los controles
negativos, consecuentemente, resultó negativa para el grupo de virus
Tobamovirus. Posteriormente se inocularon con TMV las variedades VILLA
NARRO® y SofiMely® en etapas vegetativa y antesis; confirmando la infección
con ELISA(ADGIA®), asperjando foliarmente 360°Agrocker® (3.33mL L-1) con
frecuencias de tres y siete días a partir de dicha confirmación. Se incluyeron
testigos inoculados con TMV sin aspersión de 360°Agrocker y testigos sin
inoculación ni aspersión foliar. Después de la primera aplicación foliar se
tomaron cuatro lecturas de ELISA a intervalos de 15 días cada una por etapa
inoculada, incluyendo testigos. Se analizó estadísticamente altura de planta
(AP), número de hojas (NH), monitoreo del inóculo (DO), peso fresco (PFP),
peso seco de planta (PSP), peso de fruto (PF), longitud (LF) y diámetro de fruto
(DF), número de lóculos por fruto (NL), Número de semillas por fruto (NS),
rendimiento por planta (kg PL-1), utilizando un análisis de varianza con diseño
completamente al azar y un análisis multivariado que incluyó un análisis de
componentes principales (CP) y conglomerados (AC) (Minitab.16.0); en este
último, se realizaron análisis de varianza y pruebas de Tukey (P=0.05) para los
grupos formados (SAS 9.1). Los resultados evidencian diferencias altamente
significativas (P=0.01) en las variables vegetativas y de fruto medidas, los
tratamientos libres de TMV resultaron estadísticamente superiores en todas la
variables medidas observándose en el genotipo VILLA NARRO® el mayor peso
de fruto (220.66 g) y rendimiento por planta (5.22 kg Pl-1), el genotipo SofiMely®
fue el que presentó la mayor altura de planta (213.5 cm). La inoculación del
TMV en etapa vegetativa afectó drásticamente la altura de planta en los dos
genotipos, 34.6 % en el genotipo VILLA NARRO® y 43.8% en el genotipo
SofiMely® y, así como el número de hojas 29.95 y 26.08 %, el peso fresco de
planta en 51.31 y 51.86 %, el peso seco de planta en 52.82 y 55.05 %, el peso
de fruto en 70.40 y 71.63 %, el rendimiento por planta en 76.25 % y 74.83 % y
el número de semillas por fruto en 67.46 y 65.81 % respectivamente. La
aplicación de 360°Agrocker cada tres días a una dosis de 3.33 mL L-1 en los
dos genotipos de tomate VILLA NARRO® y SofiMely®, inoculados en etapa
vegetativa, registró resultados potenciales, considerando que evitó hasta un
82.8 y 78.6 % la disminución de la altura de plantas, 85.68 y 84.5 % el número
de hojas, 85.55 y 86.39 el peso fresco de planta, 93.62 y 92.65 % el peso seco
de planta, 45.79 y 48.45 % el peso de fruto, 70.5 y 75.06 % el rendimiento por
planta y el 49.03 y 52.89 % el número de semillas respectivamente. En los
análisis multivariados, los resultados evidencian diferencias altamente
significativas entre grupos (P=0.001**) para las variables evaluadas. En el
primer componente se observó un contraste entre la absorbancia de la prueba
ELISA (DO) y el resto de las variables, teniendo mayor impacto sobre las
variables NL y LF, correlacionándose negativamente y altamente significativa
con valores de r=-0.959** y r=-0.956**, respectivamente. En el segundo
componente se observó una separación entre las variables PFP, PSP y
rendimiento e indicando una correlación positiva y altamente significativa de
rendimiento con PFP y PSP con valores de r=0.928** y r=0.878**,
respectivamente. En los tratamientos inoculados en etapa vegetativa y con
aplicación de 360°Agrocker cada tres días a una dosis de 3.33 mL L-1, se
observó una reducción máxima del inóculo en 26.73 y 19.38% en los genotipos
VILLA NARRO® y SofiMely®, respectivamente. En este estudio, 360°Agrocker
constituido por extractos vegetales nanoparticulados, tuvo la capacidad de
disminuir la cantidad de partículas virales activas pero no las redujo a niveles no
detectables. De igual manera, se evaluó la presencia de TMV en semilla de
cada tratamiento, desinfestando la semilla con hipoclorito de sodio al 10%,
utilizando 30 semillas por tratamiento y obteniendo lecturas de absorbancia de
la placa de ELISA en un espectrofotómetro, se aplicó un análisis estadístico con
un diseño completamente al azar (SAS 9.1). Por último, se evaluó la
transmisión de TMV de semilla-plántula, se sembraron en charolas de
polietileno 20 semillas por tratamiento, previamente desinfestadas con
hipoclorito de sodio al 10%, y se trasplantaron en vasos; observándose
sintomatología de TMV y corroborando con la prueba ELISA, se obtuvieron
porcentajes de transmisión. Los resultados fueron los siguientes: la presencia
de TMV en la semilla, evidencian diferencias altamente significativas (P=0.01),
todos los tratamientos exceptuando las plantas testigos libres de TMV,
resultaron positivos al virus en la prueba de ELISA en semilla, en la prueba de
transmisión de TMV de semilla-plántula, el genotipo VILLA NARRO® presentó
una transmisión del 45% de TMV, sólo cuando se inoculó el virus en etapa
vegetativa y en el genotipo SofiMely® del 55 y 40 % de transmisión de TMV,
cuando se inoculó en etapa vegetativa y en antesis, respectivamente. En ambos
genotipos los tratamientos con aplicación de 360°Agrocker®, resultaron en 0%
de transmisión de TMV" "TMV causes serious damage and crop losses of 11 to 80%, particularly under
greenhouse, where average annual losses of up to one million dollars are
calculated. The objective of the study was to evaluate the bipesticide potential of
360°Agrocker® against the pathogenesis caused by TMV in different
phenological stages in cultivated tomato. This study was carried out in the
Univesridad Autónoma Agraia Antonio Narro, starting first by evaluating the
phytosanitary quality of seed of two genotypes to be used in the research, VILLA
NARRO® and SofiMely®. These were analyzed microbiologically to detect
phytopathogens in seed, in papa-dextrose-agar (PDA) and malt salt agar (MSA)
medium. Phytopathogenic bacteria genera were identified using Gram stain,
corroborating with the Ryu test and semi-selective medium such as CNS and
Yeast-Dextrose-Calcium (YDC). The ELISA serological test (AGDIA®) was
applied to detect TMV in seeds. A completely random design with a factorial
arrangement was applied to analize de fungi data. Significant differences were
found between the number of fungi detected (P=0.05*), including genera such
as: Fusarium spp. Alternaria spp. Penicillium spp. Aspergillus spp. and
Cladosporium spp., whose incidence was 15.83, 7.5, 3.33, 4.16 and 1.66%
respectively. No significant differences were found between genotypes (P=0.91)
and culture media (P=0.07). PDA developed bacterial strains potentially
phytopathogenic yellow and orange in MSA only orange, which were associated
with the genera Xanthomonas (0.66%) and Clavibacter (0.33%) respectively; the
incidence of bacteria was low (0.74%). The absorbance readings of the
spectrophotometer, in the seed samples of the ELISA test, were lower than
those of the negative controls, consequently, it was negative for the
Tobamovirus virus group. Subsequently, the VILLA NARRO® and SofiMely®
varieties were inoculated with TMV in vegetative and anthesis stages;
confirming infection with the ELISA (ADGIA®) technique, 360°Agrocker® was
foliar sprayed (3.33 mL L-1) every three or seven days after the ELISA
confirmation. Controls inoculated in vegetative and anthesis stages without
spraying and controls without inoculation and foliar spray were included. After
the first foliar application, four ELISA readings were taken at intervals of 15 days
each per inoculated stage, including the controls. Plant height (AP), number of
leaves (NH), ELISA readings (LE), fresh weight (PFP) and dry plant weight
(PSP), fruit weight (PF), length (LF) and fruit diameter (DF), number of locules
per fruit (NL), Number of seeds per fruit (NS), yield per plant (kg PL-1), were
statistically analyzed, using multivariate analysis that included a principal
component analysis and conglomerates (Minitab 16.0). In this last, analysis of
variance and Tukey Tests (P=0.05) were performed, for the established groups
(SAS 9.1). The results showed highly significant differences (P=0.01) in the
vegetative and yield component variables, the TMV-free treatments were
statistically superior in all the variables measured, observing in the VILLA
NARRO® genotype the highest fruit weight (220.66 g) and yield per plant (5.22
kg Pl-1), and the SofiMely® genotype was the one with the highest plant height
(213.5 cm). Inoculation of TMV in the vegetative stage dramatically affected
plant height in the two genotypes, reductions of 34.6% in the VILLA NARRO®
genotype and 43.8% in the SofiMely® genotype, as well as in the number of
leaves 29.95 and 26.08%, the fresh plant weight in 51.31 and 51.86%, the dry
weight of the plant in 52.82 and 55.05%, the fruit weight in 70.40 and 71.63%,
the yield per plant in 76.25% and 74.83%, and the number of seeds per fruit in
67.46 and 65.81%, respectively. In multivariate analyzes, the results showed
highly significant differences between groups (P=0.001**) for the variables
evaluated. In the first component, a contrast was observed between the
absorbance of the ELISA test (DO) and the rest of the variables, having a
greater impact on the NL and LF variables, correlating negatively and highly
significant with values of r=-0.959** and r=-0.956**, respectively. In the second
component a separation between variables (PFP, PSP) and performance was
observed, indicating a positive and highly significant correlation of yield (kg
Plant-1) with PFP and PSP, with values of r=0.928** and r=0.878**, respectively.
In the inoculated treatments in the vegetative stage and with application of
360°Agrocker every three days at a dose of 3.33 mL L-1, a maximum reduction
of the inoculum was observed, 26.73 and 19.38% in the VILLA NARRO® and
SofiMely® genotypes, respectively. In this study, 360°Agrocker® constituted by
nanoparticulate plant extracts, had the ability to decrease the amount of active
viral particles but did not reduce them to undetectable levels. Similarly, the
presence of TMV in seed of each treatment was evaluated, disinfestating the
seed with 10% sodium hypochlorite, using 30 seeds per treatment and obtaining
absorbance readings of the ELISA plate in a spectrophotometer, a statistical
analysis was applied with a completely design randomly (SAS 9.1). Finally, the
transmission of TMV from seed-seedling was evaluated, 20 seeds were treated
in polyethylene trays per treatment, previously disinfected with 10% sodium
hypochlorite, and transplanted into vessels; observing TMV symptoms and
corroborating with the ELISA test, transmission percentages were obtained. The
results were as follows: the presence of TMV in the seed, evidences highly
significant differences (P=0.01), all treatments except the TMV-free controls
were positive for the virus in the seed ELISA test, in the seed-seedling
transmission, the VILLA NARRO® genotype presented a 45% transmission of
TMV, only when the virus was inoculated in the vegetative stage, and in the
SofiMely® genotype of 55 and 40% TMV transmission, when it was inoculated in
vegetative and anthesis stages, respectively. In both genotypes, treatments with
360°Agrocker® application resulted in 0% TMV transmission"
