RESUMEN: Se realizaron dos tratamientos térmicos a la aleación Ti6Al4V, uno por debajo
(800ºC) y otro por encima (1050ºC) de la temperatura de transformación a fase beta
(980±20ºC) variando la velocidad de enfriamiento (temple, normalizado y recocido). Se
obtuvieron dos microestructuras principales después del tratamiento térmico: globular y
laminar. El análisis por DRX mostró que las aleaciones están constituidas principalmente
de fase alfa y una pequeña parte por fase beta y fase martensítica para la aleación
Ti6Al4V1050+WQ. Las aleaciones térmicamente tratadas fueron ensayadas in vitro en una
solución fisiológica tipo Hank a 37ºC, pH 7.40±0.05 durante 7 días. Se observó el
comportamiento electroquímico a través de la evolución del potencial de circuito abierto
durante 600 s y por Espectroscopía de Impedancia Electroquímica. Las aleaciones
Ti6Al4V800+AC y Ti6Al4V1050+AC mostraron un comportamiento similar al de la aleación
Ti6Al4V sin tratamiento térmico. Con el propósito de observas la adhesión y proliferación
celular, así como los posibles cambios morfológicos de las células debido a los cambios
microestructurales se realizaron ensayos in vitro en cultivo celular (DMEM al 10% de FBS)
sembrando sobre las superficies metálicas 10,000 células osteoblasto Saos-2 y
fibroblastos 9L29, durante 7 días de inmersión a 37ºC, pH 7.40, 5%CO2 y 100% de
humedad relativa. En presencia de células osteoblasto Saos-2, los diagramas de
impedancia mostraron sistemas similares, sin embargo conforme el tiempo de inmersión
fue mayor, se observó un incremento en los valores de impedancia para las aleaciones
Ti6Al4V800+AC y Ti6Al4V1050+AC. Por otro lado, en presencia de células fibroblasto 9L29, los
diagramas de impedancia mostraron un sistema estable para las tres aleaciones conforme
el tiempo de inmersión fue mayor, observándose valores impedancia similares para las
aleaciones Ti6Al4V800+AC y Ti6Al4V1050+AC y mayores para la aleación Ti6Al4V sin
tratamiento térmico. Los análisis de XPS, mostraron que el comportamiento de los
materiales en el medio de cultivo se debe a la formación de TiO2 durante su esterilización
y en menor grado a la adsorción de proteínas y/o células sobre el óxido. La adhesión de
las células fue caracterizada por SEM/EDX, dónde en las aleaciones Ti6Al4V sin
tratamiento térmico y Ti6Al4V800+AC se observan células osteoblasto con morfología
poligonal y alargada, mientras que para la aleación Ti6Al4V1050+AC la morfología es más
esférica y éstas se acumular en ciertas zonas de la superficie, con mayor recubrimiento
sobre la aleación Ti6Al4V sin tratamiento térmico. Respecto a las células fibroblasto, la
adhesión y proliferación celular es mayor que para las células osteoblasto, observándose
células interconectadas con morfología poligonal. Adicionalmente, en la superficie de las
aleaciones Ti6Al4V sin tratamiento térmico y Ti6Al4V800+AC se observaron pequeñas islas,
mientras que para la aleación Ti6Al4V1050+AC esto no se observa. La adhesión,
proliferación y morfología celular, son dependientes de la microestructura de los
materiales. El análisis por EDX de estas superficies permitió identificar las señales de los
elementos Ti y O debido a la formación del óxido y las señales de C, N y O, relacionado
con la formación de compuestos orgánicos provenientes de la matriz extracelular, tanto en
presencia de células osteoblasto como fibroblasto. Sin embargo, en la biopelícula óxido
pasivo/fibroblastos se identificaron los elementos Na, Cl, S, P que modificaron la
adhesión, proliferación y morfología celular de los fibroblastos, en tanto que para la
biopelícula óxido pasivo/osteoblastos solo se identificó P para la aleación Ti6Al4V800+AC.
Se detectó Ca sobre la biopelícula óxido pasivo/osteoblastos de las aleaciones
Ti6Al4V800+AC (<0.22w%) Ti6Al4V1050+AC (<0.33w%) y solo sobre la biopelícula óxido
pasivo/fibroblastos para la aleación Ti6Al4V1050+AC (0.75w%). A partir de estos resultados,
se propone que la adhesión de las células es más favorable sobre el TiO2 formado en
regiones discretas donde se tiene la fase α (HCC); esto podría deberse a que la formación
de la película de óxido es más defectuosa y/o se encuentra mayormente hidratada,
facilitando la adhesión celular. La osteointegración de las aleaciones Ti6Al4V sin
tratamiento térmico, Ti6Al4V800+AC y Ti6Al4V1050+AC se evaluó mediante ensayos in vivo
implantado estos materiales en animales experimentales (ratas tipo Wistar) durante 15, 30
y 60 días. Los resultados muestran que todas las superficies se osteointegraron desde los
primeros 15 días, observándose hueso maduro alrededor del implante a los 60 días.
ABSTRACT: Heat treatments were performed on the Ti6Al4V alloy, one below (800°C) and one
above (1050°C) from the transformation temperature to beta phase (980±20°C), and
varying the cooling rate (water quenching, air cooling and furnace cooling). Two main
microstructures were obtained after heat treatments: globular and lamellar. X-Ray
Diffraction (XRD) analysis showed that the alloys consist mainly of alpha phase and a
small part of beta phase and martensitic phase was identify for the Ti6Al4V1050+WQ alloy.
The heat-treated alloys were tested in vitro on a physiological solution Hank at 37 ° C, pH
7.40 ± 0.05 for 7 days. The electrochemical behavior of these alloys was observed through
the evolution of the Open Circuit Potential (OCP) for 600s and using Electrochemical
Impedance Spectroscopy (EIS) ac technique. Ti6Al4V800+AC and Ti6Al4V1050+AC alloys
showed a similar behavior regarding Ti6Al4V as received alloy. In order to observe cell
adhesion and cell proliferation, as well as possible morphological changes of the cells due
to microstructural changes, in vitro cell culture assays (10% DMEM of FBS) were
performed by seeding on metal surfaces 10,000 Saos-2 osteoblast cells and 9L29
fibroblasts (in separate experiments) for 7 days at 37 ° C, pH 7.40, 5% CO 2 and 100%
relative humidity. In presence of Saos-2 osteoblast cells, the impedance diagrams showed
a similar electrochemical behavior, however, as the immersion time was higher, an
increase in the impedance values was observed for Ti6Al4V800+AC and Ti6Al4V1050+AC alloys.
On the other hand, For 9L29 fibroblast cells, the impedance diagrams showed a stable
system for the three alloys and according to the immersion time was higher, similar
impedance values were observed for the Ti6Al4V800+AC and Ti6Al4V1050+AC alloys and higher
for the Ti6Al4V as received alloy. XPS analysis showed that the behavior of the materials
in the culture medium is mainly due to the formation of TiO2 after its sterilization and to a
lesser extent the adsorption of proteins and/or cells on the passive oxide. Cell adhesion
was characterized by SEM/EDX, where on the Ti6Al4V as received and Ti6Al4V800+AC
alloys, osteoblast cells with polygonal and elongated morphology were observed, whereas
on the Ti6Al4V1050+AC alloy, the morphology was more spherical and these were
accumulate in certain areas of the surface, with greater coating on the Ti6Al4V as received
alloy. Regarding fibroblast cells, cell adhesion and proliferation is greater than for
osteoblast cells, with interconnected cells and polygonal morphology being observed.
Additionally, on the Ti6Al4V as received surface and Ti6Al4V800+AC alloys, small cell islands
were observed, whereas on the alloy Ti6Al4V1050+AC it was not observed. Cell adhesion,
proliferation and morphology were dependent on the microstructure of the materials. EDX
analysis of these surfaces allowed us to identify the signals of the elements Ti and O due
to the formation of the oxide and the signals of C, N and O related to the formation of
organic compounds from the extracellular matrix, both for osteoblast cells as fibroblasts
cells. However, in the biofilm passive oxide/fibroblast cells, Na, Cl, S, P elements that
modified cell adhesion, proliferation and cell morphology of the fibroblasts, whereas for the
biofilm passive oxide/osteoblast cells only P was identified on the Ti6Al4V800+AC surface
alloy. Ca was detected on the biofilm of passive oxide/osteoblasts cells on the
Ti6Al4V800+AC (<0.22w%) Ti6Al4V1050+AC (<0.33w%) surface alloys and on the biofilm
passive oxide/fibroblast cells on the Ti6Al4V1050+AC alloy (0.75w%). From these results, it is
proposed that the cell adhesion is more favorable on the TiO2 formed in discrete regions
where the α phase (HCC) is present; it could be due to the formation of the oxide film
being more defective and/or being largely hydrated, facilitating cell adhesion.
Osseointegration of Ti6Al4V as received, Ti6Al4V800+AC and Ti6Al4V1050+AC alloys was
evaluated by in vivo assays implanted in experimental animals (Wistar rats) for 15, 30 and
60 days. The results show that all surfaces show osseointegration from the first 15 days,
with mature bone being observed around the implant at 60 days.
