Modulación de la vía de PI3/AKT en cultivos celulares de retina : rol del óxido nítrico y vías de señalización mediadas por calcio-calmodulina

Resumen
El óxido nítrico presenta efectos en la supervivencia de célular neuronales in vitro sometidas a efectos de muerte por degeneración a través de la via clásica del óxido nítrico. El S-Nitroso-N-acetil-penicilamina (SNAP) o L-Arginina ()L-ARG) son buenos agentes que generan óxido nítrico intracelular y analiza la fosforilación de la proteína quinasa citoplasmática AKT y su consiguiente translocación nuclear en su forma activa para los sitios fosforilados de la Ser-473 y Thre-308. Los resultados muestran que SNAP y L-ARG inducen la activación de AKT vía la ruta clásica del óxido nítrico y su consiguiente migración nuclear del citoplasma hacia el núcleo tanto en tres tipos celulares en cultivo in vitro de neuronas primarias de retina PC12 y SN56. Esos efectos son bloqueados cuando son tratadas con diferentes inhibidores de la PKG, AKT, PI3K, CaMKII, Calmodulina. Brasil. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) Tesis
