Genotoxicidad de cloroquina en mesocricetus auratus “hámster”

El presente estudio tuvo como finalidad evaluar la genotoxicidad de cloroquina mediante el análisis de micronúcleos y alteraciones cromosómica en células de médula ósea y el ensayo de anomalías en los espermatozoides de Mesocricetus auratus. Se trabajo con un control negativo (agua destilada estéril), un control positivo (50 mg/Kg/pc ciclofosfamida) y tres grupos experimentales tratados con cloroquina a razón de 250, 300 y 350 mg/Kg. p.c. Los resultados muestran que el índice de genotoxicidad de los grupos experimentales (5.65±0.9, 6.25±0.72, 6.44±0.39) y el control positivo (6.55±0.49) presentan diferencias significativas en relación al control negativo (1.23±0.12), presentándose variaciones en número y tamaño del micronúcleo. Así mismo, las alteraciones cromosómicas también se presentan diferencias significativas entre y los grupos experimentales (2.0±0.32, 2.76±0.67, 3.16±0.38) y control positivo (4.0±0.5) en relación al control negativo (0.96±0.38), presentándose fracturas en una o ambas cromátidas del cromosoma y cromosomas dicéntricos. El análisis de espermatozoides, revela que la frecuencia de espermatozoides con cabezas anormales presenta diferencias significativas entre los grupos de tratamiento (2.54±011, 4.02±0.19, 3.42±0.25) y control positivo (3.62±0.3) en relación al control negativo (1.82±0.25). Se encontró espermatozoides con dos colas, dos cabezas y espermatozoides plegados. Se concluye que Cloroquina es genotóxica para células de médula ósea e induce alteraciones en la morfología de los espermatozoides de M. auratus y puede representar un riesgo para la salud humana
