Uso de delineamiento experimental para el estudio de un proceso de granulación húmeda en Reómetro de Torque

La granulación es una estrategia utilizada para viabilizar la producción de comprimidos cuando la dosis de un fármaco es bastante elevada y/o sus propiedades de flujo imposibilitan la utilización de la compresión directa. El presente trabajo tuvo por objetivo utilizar un delineamiento factorial fraccionado para estudiar la influencia de los parámetros empleados en la reometría de torque y su impacto en los gránulos producidos. Para estudiar el proceso de granulación húmeda, se desarrolló la técnica de estudio de la reología de los materiales por medio del torque, siendo el reómetro de torque (Mixer Torque Rheometer - MTR) una importante herramienta para estudiarla. En el presente trabajo, se evaluaron los perfiles reológicos de mezclas de celulosa microcristalina PH 101 con polivinilpirrolidona (PVP K30), planificándose los ensayos por medio del delineamiento experimental de tipo factorial fraccionado, con 2 puntos centrales, teniendo como factores independientes el volumen de líquido de granulación (2, 4 y 8 mL), tiempo de mezcla (30, 60 y 90 s) y la velocidad de agitación (25, 50 y 75 rpm), cada uno variado en tres niveles y utilizando un total de muestra de 15 gramos. Los resultados obtenidos en el MTR se sometieron a la evaluación estadística, que a su vez indicó la posible influencia de la velocidad de rotación de las hélices en el perfil reológico de las mezclas. A partir de esta información, se seleccionaron tres ensayos (Run 2, Run 4 y Run 7) para ser producidos con diferentes velocidades de agitación en mezclador orbital planetario, siguiendo la razón líquido / sólido suministrada por el MTR para cada una de ellas. Después del proceso de humidificación, los granulados fueron secados en la estufa y, a continuación, se caracterizaron según la densidad verdadera, densidad compactada, índice de Carr y proporción de Hausner para propiedades de flujo, friabilidad, distribución granulométrica y morfología de las partículas. Los granulados presentaron características similares en cuanto a distribución granulométrica, densidades y morfología. Sin embargo, aquellos producidos con menor velocidad de agitación presentaron mayor friabilidad. Se concluyó que la velocidad de rotación de las hélices en el MTR influye significativamente en el perfil reológico de la mezcla que, a su vez, tuvo impacto también en la resistencia de los gránulos obtenidos.
