Cinética da infecção aguda pelo Toxoplasma gondii e alterações histopatológicas no duodeno de ratos

Uma forma comum de transmissão é a ingestão de alimentos contaminados com oocistos. O Toxoplasma gondii precisa atravessar a barreira intestinal, invadindo células epiteliais, alcançando a parede intestinal e se disseminando pelo organismo. Objetivamos analisar a parede intestinal e as células do epitélio do duodeno de ratos infectados por Toxoplasma gondii durante diferentes durações de infecção aguda. Ratos foram separados em grupos inoculados por via oral com oocistos de Toxoplasma gondii por 6 horas (G6), 12 horas (G12), 24 horas (G24), 48 horas (G48), 72 horas (G72), 7 dias (G7d) e 10 dias (G10d), e em grupo controle não infectado (GC). Aos 60 dias de idade, esses ratos foram submetidos à eutanásia e os duodenos processados histologicamente para análise da parede intestinal e de células epiteliais. Nos grupos infectados foi observado aumento na largura de vilos e criptas; redução na altura de enterócitos; aumento no menor diâmetro e redução no maior diâmetro do núcleo de enterócitos. Ocorreu aumento na quantidade de células caliciformes que secretam muco neutro no G6, G12 e G7d e das que secretam muco ácido no G7d e G10d, além de aumento no número de linfócitos intraepiteliais no G48. Houve ainda atrofia da tela submucosa, da túnica muscular e da parede total nos grupos infectados. Conclui-se que diferentes tempos de infecção aguda por Toxoplasma gondii causam alterações na estrutura histológica do duodeno de ratos no intuito de manter a homeostasia do animal ABSTRACT: Toxoplasma gondii is a prevalent parasite among humans and animals. A common form of transmission is through the ingestion of food contaminated with oocysts. Toxoplasma gondii must cross the intestinal barrier, invading epithelial cells, reaching the intestinal wall and spreading throughout the body. We aimed to analyze the intestinal wall and epithelial cells of the duodenum of rats infected with Toxoplasma gondii during different durations of acute infection. Rats were separated into groups inoculated orally with Toxoplasma gondii oocysts for 6 hours (G6), 12 hours (G12), 24 hours (G24), 48 hours (G48), 72 hours (G72), 7 days (G7d) and 10 days (G10d), and an uninfected control group (CG). When rats were 60-day-old, they were euthanized and the duodenum were histologically processed to analyze the intestinal wall and epithelial cells. We observed in the infected groups increase in the width of villi and crypts; reduction in the height of enterocytes; increase in the minor diameter and decrease in the major diameter of the enterocyte nucleus. An increase of goblet cells secreting neutral mucus in G6, G12 and G7d was observed as well as of goblet cells secreting acid mucus in G7d and G10d. An increased number of intraepithelial lymphocytes was seen in G48. There was atrophy of the submucosa, the external muscle layer and the total wall in the infected groups. It can be concluded that different period of acute Toxoplasma gondii infection causes changes in histological structure of the duodenum from rats in order to maintain homeostasis of the animal 55[8] f. : il.
