Ministério da Educação e Universidade Federal Rural da Amazônia Células-tronco mesenquimais (CTMs) são caracterizadas por serem células indiferenciadas
que possuem como principais características a capacidade de autorenovação, alto potencial de
proliferação e diferenciação em diversos tecidos. São facilmente isoladas in vitro,
apresentando morfologia semelhante a fibroblastos e devem apresentar a capacidade de
realizarem in vitro a diferenciação condrogênica, osteogênica e adipogênica, assim como
serem positivas para CD105, CD73 e CD90 e negativas para CD45, CD34, CD79a. O
presente estudo tem como objetivo avaliar o potencial das células-tronco mesenquimais
derivadas de tecido adiposo de Aotus azarai infulatus KUHL-1820, oriundos do Centro
Nacional de Primatas (CENP)- Ananindeua, PA. Acredita-se que tal espécie animal possui
características que lhes permitem ser um excelente modelo biológico para estudos com fins de
terapia celular, objetivou também analisar a curva de crescimento celular, porcentagem da
viabilidade celular, antes e após a criopreservação, realizar análises morfológicas, testes
imunocitoquímicos e a análises do cariótipo. As células foram isoladas por digestão
enzimática com colagenase tipo IV e cultivadas in vitro. Os resultados mostraram que as
células-tronco mesenquimais derivadas de tecido adiposo possuem elevada capacidade de
proliferação se expandindo rápido nas garrafas de cultivo, apresentaram viabilidade em média
acima de 90% e não apresentou diferença significativa quando se comparou células
submetidas à criopreservação com células não criopreservadas. Apresentaram morfologia
semelhante a fibroblastos e uma cinética de crescimento esperada para esse tipo de célula,
foram positivas para CD105, CD73, e CD90 e negativa para CD34, CD79a e CD45. Na
análise cariotípica foi observado um rearranjo em heterozigose no par 3, no qual um dos
cromossomos sofreu uma fissão cêntrica, originando dois acrocêntricos, sugerindo que o
mesmo é uma alteração constitucional do animal analisado, sugerindo que o mesmo seja
híbrido entre dois citótipos diferentes. As células responderam de maneira positiva para a
diferenciação adipogênica, condrgênica e osteogênica, comprovando sua
multipotencialidade.Conclui-se, através dos resultados obtidos, que os testes básicos
necessários para que uma célula seja considerada célula-tronco mesenquimal foram positivos
nestes experimento, induzindo que a espécie Aotus azarai infulatus pode servir como
excelente modelo biológico em estudos utilizando células-tronco mesenquimais e terapia

Derived Mesenchymal Stem Cells of Aotus azarai infulatus KUHL-1820, obtained from the
National Primate Center (CENP) - Ananindeua, PA. It is believed that this specie has
characteristics that allows them to be an excellent biological model for studies of cellular
therapy, also examining the cell growth curve, the percentage of cell viability before and after
cryopreservation, performing morphological analysis, immunocytochemical tests and
karyotype analyzes. The Cells were isolated by enzymatic digestion with type IV collagenase
and cultured in vitro. The results showed that Adipose-Derived Mesenchymal Stem Cells has
high proliferation capacity expanding fast in bottles of cultivation, showed average viability
above 90% and was not significantly different when compared with cells subjected to
cryopreservation and not cryopreserved cells . Showed fibroblast-like morphology and growth
kinetics expected for this type of cells. Were positive for CD105, CD73, and CD90 and
negative for CD34, CD45 and CD79a. In karyotype analysis showed a rearrangement
heterozygous at par 3, in which one of the chromosomes underwent a centric fission, yielding
two acrocentric, suggesting that it is a constitutional alteration of the analyzed animal,
suggesting that it is hybrid between two different cytotypes. The cells responded positively to
the adipogenic, chondrogenic and osteogenic differentiation. The results, showed that the
basic tests required for a cell to be considered mesenchymal stem cells were positive in these
experiments, leading to the Aotus azarai infulatus may be an excellent biological model in
studies using mesenchymal stem cells and cellular therapy.
