Resíduos de antibióticos em leite: limite de detecção por espectroscopia óptica

Milk, rich in various nutrients, plays an important role in human nutrition. Quality is closely linked to the effectiveness of the handling process. The presence of antibiotic residues in bovine milk causes serious losses in the production of cheeses, yogurts and curds. The ingestion of these residues threatens the health of the consumer favoring microbial resistance and hypersensitivity. Thus, the objective of this study is to test the application of Raman and Infrared spectroscopy techniques, which stand out as analytical tools of great potential, since they provide fast, quantitative, qualitative and reliable results, requiring little or no pretreatment of the samples. The conformation and vibrational spectrum of the antibiotic molecules Penicillin and Ampicillin were investigated by means of a computational analysis with the aid of a B3LYP hybrid functional at the DFT theory level. As of Resolution 53 of October of the National Agency of Sanitary Surveillance (ANVISA), Brazil follows the Maximum Residue Limits (MRLs) established by Mercosur. The analyzed milk was separated into two portions, the genuine and the control being that the latter was simulated the antibiotic contamination following the MRLs. Pre- or post-treatment of the samples was not required for the analyzes. The results indicate that this methodology can be used in the future to identify the type and quantification of the antibiotic that may be present in bovine milk. O leite, rico em vários nutrientes, desempenha um papel importante na nutrição humana. Sua
qualidade está intimamente ligada a eficácia do processo de manipulação. A presença de
resíduos de antibióticos no leite bovino causa sérios prejuízos na produção de queijos, iogurtes
e coalhos. A ingestão desses resíduos ameaça a saúde do consumidor favorecendo a resistência
microbiana e a hipersensibilidade. Assim, o objetivo deste estudo é testar a aplicação de técnicas
de espectroscopia Raman e Infravermelho, que se destacam como ferramentas analíticas de
grande potencial, uma vez que proporcionam resultados rápidos, quantitativos, qualitativos e
confiáveis, exigindo pouco ou nenhum pré-tratamento das amostras. Investigou-se por meio de
análise computacional a conformação e o espectro vibracional das moléculas dos antibióticos
Penicilina e Ampicilina, com auxílio de um funcional híbrido B3LYP no nivel de teoria DFT.
A partir da Resolução 53 de outubro da Agência Nacional de Vigilância Sanitária (ANVISA),
o Brasil segue os Limites Máximos de Resíduos (LMR) estabelecidos pelo Mercosul. O leite
analisado foi separado em duas porções, o genuíno e o controle sendo que nesta última foi
simulada a contaminação por antibiótico seguindo os LMR. Não foi necessária a realização de
pré ou pós-tratamento das amostras para as análises. Os resultados obtidos indicam que esta
metodologia possa ser utilizada futuramente na identificação do tipo e na quantificação do
