Antioxidant and anti-glycation capacities of some medicinal plants and their potential inhibitory against digestive enzymes related to type 2 diabetes mellitus

Some medicinal plants are able to providing antioxidant compounds that act against
oxidative stress and protein glycation and others that are capable of inhibiting the
catalysis of digestive enzymes and thus possibly contributing to the reduction of
postprandial hyperglycemia and hyperlipidemia. Our objective was to investigate the
antioxidant and anti-glycation activities of some medicinal plants and their potential
inhibitory against α-amylase, α-glucosidase and pancreatic lipase activities. Based on
the ethnobotanical researches carried out by academic studies conducted at the
Federal University of Uberlandia, ten plants traditionally used in the treatment of type-
2 diabetes mellitus were selected. Ethanol (ETOH) and hexane (Hex) extracts of
specific parts of these plants were used in enzymatic assays to evaluate their
inhibitory potential against α-amylase, α-glucosidase and lipase, as well as their
antioxidant (DPPH, ORAC and FRAP) and anti-glycation (BSA/fructose model)
capacities. The results indicate that ETOH extract of four of the ten analyzed plants
exhibited more than 70% of antioxidant and anti-glycation capacities, and α-amylase
and lipase inhibitory activities; no extract was able to inhibit more than 40% the α-
glucosidase activity. The ETOH extracts of Bauhinia forficata and Syzygium cumini
inhibited α-amylase, while the ETOH extracts of B. forficata, Matricaria recutita and
Echinodorus grandiflorus inhibited lipase. In addition, the ETOH extracts of B.
forficata, S. cumini, M. recutita and E. grandiflorus presented higher antioxidant
capacity, respectively. and anti-graft activity. ETOH extracts from four of the ten
species popularly cited for treatment of type 2 diabetes mellitus showed promising
antioxidant and anti-glycating properties as well as ability to inhibit α-amylase and
lipase digestive enzymes. The partitions of B. forficata and S. cumini showed
prominent results in all trials. Thus, our results open new possibilities for others

enzimas digestivas e assim, possivelmente, contribuir para a redução de
hiperglicemia e hiperlipidemia pós-prandial. O objetivo deste trabalho foi investigar
as atividades antioxidantes e antiglicantes de algumas plantas medicinais e seu
potencial inibitório contra as atividades da α-amilase, α-glucosidase e lipase
pancreática. Considerando pesquisas etnobotânicas realizadas na Universidade
Federal de Uberlândia, foram selecionadasn dez plantas tradicionalmente utilizadas
como tratamento complementar ao diabetes mellitus tipo 2. Extratos de etanol
(ETOH) e hexano (Hex) de partes específicas dessas plantas foram utilizados em
ensaios enzimáticos para avaliar seus potenciais de inibição contra α-amilase, α-
glicosidase e lipase, bem como sua capacidade antioxidante (DPPH, ORAC e FRAP)
e antiglicante (BSA / frutose). Os resultados indicam que o extrato ETOH de quatro
das dez plantas analisadas apresentou mais de 70% de capacidade antioxidante e
antiglicante, bem como alta inibição das enzimas α-amilase e lipase; nenhum extrato
foi capaz de inibir mais de 40% da atividade da α-glicosidase. Os extratos ETOH de
Bauhinia forficata e Syzygium cumini inibiram a α-amilase, enquanto os extratos
ETOH de B. forficata, Matricaria recutita e Echinodorus grandiflorus inibiram a lipase.
Além disso, os extratos ETOH de B. forficata, S. cumini, M. recutita e E. grandiflorus
apresentaram, respectivamente, maior capacidade antioxidante. e atividade
antiglicante. Os extratos de ETOH de quatro das dez espécies popularmente citadas
para o tratamento complementar do diabetes mellitus tipo 2 mostraram propriedades
antioxidantes e antiglicantes promissoras, bem como capacidade de inibir as
enzimas digestivas α-amilase e lipase. As partições de B. forficata e S. cumini
apresentaram resultados proeminentes em todos os ensaios. Assim, nossos
resultados abrem novas possibilidades para outros estudos, a fim de avaliar o
potencial antidiabético dessas plantas medicinais.
