Arquivo retido, a pedido da autora, até janeiro de 2024. Insect meal is a promising source, rich in protein, lipids and other nutrients and can be used as
a substitute for animal or vegetable protein. However, the lack of chitinolytic enzymes in the
gastrointestinal tract of some fish, necessary for chitin breakdown, causes the non-utilization
of this raw material. Confronting this, the application of nutritional or metabolic programming
is interesting, since exposure during the animal’s development phase to a stimulus of
endogenous or exogenous origin can lead to long-term structural or functional changes.
Therefore, based on the nutritional programming concept in mammals, this project evaluated
the ability of tilapia fingerlings to use diets with a high content of insect meal on the lipid
profile and intestine oxidative stress after consuming giant mealworm (TG) during
larviculture. To carry out the test, initial preparation in the larval stage with the presence or
not of SW meal was necessary. After 40 days, biometry was performed, with male fingerlings
(3.1 ± 0.08 g average weight) reallocated in groups in a 2x3 factorial design with or without
TG and experimental diets (0, 150 and 300 g/Kg TG) fed twice a day for 60 days. After those
60 days, they were euthanized and fillet and liver were collected for fatty acid analysis (FA);
plasma for triglycerides, protein, NAG, cholesterol and total protein analysis and finally, the
midgut for oxidative stress. Regarding FA in fillet and liver, it was observed that the diet with
the TG inclusion increased SAFA and MUFA, this not being true for PUFA. In the liver,
similar to PUFA, HUFA decreased with the TG inclusion. Only in the liver it was possible to
visualize the effect of nutritional programming, this being reflected in the parameters C16:0,
C20:4n6, C22:6n3, n3, HUFA, SAFA, n3/n6 ratio a decrease and an increase over C20:3n3
and n6/n3 ratio, however the observations are restricted to the 0 diet. The FA profile found in
both tissues reflected negatively on the nutritional quality index, with the TG inclusion. In
plasma, only NAG, protein and triglycerides were significant for programming. NAG and
protein content were higher in the fish submitted to programming, while the triglycerides of
the 0 and 150 g/Kg diets were lower, not being true for the 300 g/Kg diet. Catalase, superoxide
dismutase and TBARS were evaluated on oxidative stress, however they did not show
significant effects. The histopathology and lipid inclusion in the liver were not significant,
except for vacuolization, that was lower in the 300 g/Kg TG inclusion diet. In conclusion,
nutritional programming and diets were able to interfere with the lipid profile and liver
vacuolization and plasma parameters. The results for the lipid profile found that the diets were
not able to interfere with the animal’s health since the 300 g/Kg TG inclusion led to a decrease
in vacuolization in the liver. We also highlighted the nutritional programming influence for
the chitin use, demonstrating the possibility of including insect meal in fish feed. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) A farinha de insetos é uma fonte promissora, rica em proteína e lipídeos, podendo ser
utilizada em substituição à proteína animal ou vegetal. No entanto, a falta de enzimas
quitinolíticas no trato gastro intestinal de alguns peixes, necessária para a quebra de quitina,
ocasiona o seu não aproveitamento. Visto isso, a aplicação da programação nutricional se
mostra interessante, uma vez que a exposição durante a fase de desenvolvimento do animal
leva a um estímulo de origem endógena ou exógena, pode levar a mudanças fisiológicas a
longo prazo. Com base neste conceito, o projeto avaliou a capacidade de alevinos de tilápia
em utilizar dietas com alto teor de farinha de insetos sobre o perfil lipídico e o estresse
oxidativo do intestino após consumir a farinha de tenébrio gigante (TG) durante a larvicultura.
Para a realização do ensaio foi realizada a programação nutricional na fase larval com a
presença ou não de TG. Após 40 dias realizou-se uma biometria, onde os alevinos machos
foram redistribuidos em delineamento fatorial 2 x 3, com ou sem TG na larvicultura e três
níveis de TG na dieta (0, 150, 300 g/Kg). Os animais foram alimentados por 60 dias. Ao final
foram sacrificados, sendo coletados filé e fígado para as análises de ácidos graxos (AG);
plasma para análise de triglicerídeos, proteína, n-acetilglucosamina (NAG), colesterol e
proteína total e por fim intestino médio para estresse oxidativo. Quanto ao AG no filé e no
fígado, observou-se que a dieta com inclusão de TG aumentou os SAFA e MUFA, não sendo
verdadeiro para o PUFA. No fígado, assim como os PUFA, os HUFA diminuíram com a
inclusão de TG. No fígado foi possível visualizar o efeito da programação, sendo refletido nos
parâmetros C16:0, C20:4n6, C22:6n3, n3, HUFA, SAFA, relação n3/n6 uma diminuição e um
aumento sobre C20:3n3 e a relação n6/n3, contudo as observações se restringem a dieta 0. O
perfil de AG encontrado em ambos tecidos refletiu negativamente no índice de qualidade
nutricional, com a inclusão de TG. No plasma somente NAG, proteína e triglicerídeos foram
significativos a programação. O teor de NAG e proteína foram maiores nos peixes submetidos
à programação, enquanto os triglicerídeos da dieta 0 e 150 g/Kg foram menores, não sendo
verdadeiro para dieta 300 g/Kg. OS parâmetros avaliados do estresse oxidativo não
demonstraram efeitos significativos. A histopatologia e a inclusão lipídica no fígado não
foram significativas, exceto pela vacuolização, que foi menor na dieta de inclusão de 300
g/Kg de TG. Em conclusão, a programação nutricional e as dietas foram capazes de interferir
no perfil lipídico, vacuolização do fígado e nos parâmetros plasmáticos. Os resultados de
perfil lipídico encontrados não foram capazes de interferir na saúde do animal uma vez que a
inclusão de 300 g/Kg TG levou a diminuição da vacuolização no fígado. E evidenciamos a
importância da programação nutricional para a utilização da quitina, demostrando a
possibilidade da inclusão de farinha de insetos.
