Glutationa modula a liberação de adenosina em cultura primária de astrócitos corticais de camundongo

Glutathione (GSH) is one of the main antioxidants in the Central Nervous System
(CNS) and a potential gliotransmitter, inducing calcium waves in the cytosol of glial cells.
Adenosine (Adn) is a neuromodulator widely expressed in the CNS and its extracellular
levels are a critical factor in determining its effect on nervous tissue. It is known that Ca2+-
dependent pathways regulate Adn release. Since GSH has the ability to induce Ca2+
waves in the cytosol of glial cells, the present work aims to investigate whether this
molecule can regulate extracellular DNA levels. To assess this, we used primary cultures
of cortical astrocytes maintained in DMEM+10% SBF in a CO2 oven (37oC, 95% O2/5%
CO2) for 12-15 days, when they reached confluence. The cells were incubated with Hank
buffer for different time intervals, after which this solution was collected and the
neurotransmitters present there were quantified by High Performance Liquid
Chromatography. Our data show that GSH induces an 80% increase in extracellular Adn
levels at two analyzed times: 5 and 20 minutes. Removal of GSH from the incubation
medium returns the Adn concentration to baseline levels. Removal of Na+ or Ca2+ from
the medium did not affect the effect of GSH. Blockade of nucleoside equilibrative
transporters by dipyridamole (10 µM) significantly decreased the levels of Adn in the
medium, but did not interfere with the action of GSH. In order to assess whether the effect
of GSH derives from an indirect modulation on the release of glutamate or GABA (two
agents described as regulators of Adn release), the quantification of these transmitters
was performed. Both were significantly increased in the presence of GSH. However,
unlike what was observed with Adn, the removal of Na+ from the incubation medium
mitigated the effect of GSH on glutamate release. The incubation of astrocytes with GABA
(50 and 100 µM) did not influence the extracellular Adn concentration in our experimental
model, ruling out a GABAergic modulation behind the effect of GSH. The evaluation of
redox agents showed that thiol compounds reproduce the effect of GSH, while the non thiol antioxidant alpha-tocopherol did not regulate extracellular Adn levels. Thus, the
present work concludes that astrocytes express a GSH-sensitive component that can be

gliais. A adenosina (Adn) é um neuromodulador amplamente expresso no SNC e os seus
níveis extracelulares são um fator crítico para determinar o seu efeito no tecido nervoso.
Sabe-se que vias dependentes de Ca2+ regulam a liberação de Adn. Uma vez que a GSH
tem a capacidade de induzir ondas de Ca2+ no citosol de células gliais, o presente
trabalho se propõe a investigar se essa molécula pode regular os níveis extracelulares
de Adn. Para avaliar isso, foram utilizadas culturas primárias de astrócitos corticais
mantidas em DMEM+10% SBF em estufa de CO2 (37oC, 95% O2/5% CO2) por 12-15
dias, quando atingiram a confluência. As células foram incubadas com tampão Hank por
diferentes intervalos de tempo, após o qual foi realizada a coleta dessa solução e os
neurotransmissores ali presentes foram quantificados por Cromatografia Líquida de Alta
Eficácia. Nossos dados mostram que a GSH induz um aumento de 80% nos níveis
extracelulares de Adn em dois tempos analisados: 5 e 20 minutos. A remoção da GSH
do meio de incubação faz a concentração de Adn retornar aos níveis basais. A remoção
de Na+ ou Ca2+ do meio não interferiu no efeito da GSH. O bloqueio dos transportadores
equilibrativos de nucleosídeos por dipiridamole (10 µM) diminuiu significativamente os
níveis de Adn no meio, mas não interferiu na ação da GSH. A fim de avaliar se o efeito
da GSH deriva de uma modulação indireta sobre a liberação de glutamato ou GABA
(Dois agentes descritos como reguladores da liberação de Adn), foi realizada a
quantificação desses transmissores. Ambos foram significativamente aumentados na
presença da GSH. No entanto, diferente do que foi observado com a Adn, a remoção de
Na+ do meio de incubação mitigou o efeito da GSH sobre a liberação de glutamato. A
incubação dos astrócitos com GABA (50 e 100 µM) não influenciou na concentração
extracelular de Adn em nosso modelo experimental, descartando uma modulação
GABAérgica por trás do efeito da GSH. A avaliação de agentes redox demonstrou que
compostos tiol reproduzem o efeito da GSH, enquanto o antioxidante não-tiol alfa tocoferol não regulou os níveis extracelulares de Adn. Desse modo, o presente trabalho
conclui que os astrócitos expressam um componente sensível à GSH que pode ser
modulado pelo seu grupamento sulfidrila.
