Os venenos de serpentes são misturas ricas em proteínas, enzimas e
peptídeos biologicamente ativos, os quais representam aproximadamente
90% dos constituintes totais e, provavelmente, são responsáveis por grande parte dos efeitos biológicos do veneno. A L-aminoácido oxidase (LAAO) é uma das enzimas encontradas nos venenos e muitos são os efeitos atribuídos a esta enzima: antimicrobiano, indutor de apoptose, agregação plaquetária, anti-câncer, dentre outros. Considerando a abrangência de ações das LAAOs, o objetivo deste trabalho foi caracterizar bioquimicamente uma LAAO isolada do veneno bruto de Bothrops jararacussu e avaliar seu potencial antibacteriano e antitumoral. O isolamento da LAAO de B. jararacussu envolveu três etapas de purificação: uma cromatografia de afinidade à concanavalina A e duas etapas de cromatografia por troca iônica em coluna MonoQ. O alto grau de pureza da proteína foi confirmado por SDS-PAGE, e a massa molecular estimada foi de aproximadamente 68 kDa. A LAAO isolada apresentou maior especificidade enzimática por L-Fen, seguida por L-Met e L-Arg. O perfil de peptídeos trípticos revelou homologia com outra LAAO já isolada de B. jararacussu, porém a sequência N-terminal obtida (ATNRNPLYYQFRRTDYFIF) mostrou 56% de identidade com essa
enzima, podendo se tratar de uma nova LAAO obtida de B. jararacussu. A
LAAO isolada apresentou atividade antimicrobiana contra bactérias Gramnegativas - Proteus vulgaris (CIM 25 μg/mL, CBM 50 μg/mL) e Citrobacter freundii (CIM 100 μg/mL, CBM 100 μg/mL) e Gram-positivas - Listeria monocytogenes (CIM 25 μg/mL), Bacillus cereus (CIM 400 μg/mL), Bacillus subtilis (CIM 100 μg/mL, CBM 200 μg/mL) e Staphylococcus aureus (CIM 50 μg/mL, CBM 50 μg/mL). Para esta última cepa a LAAO mostrou ser mais efetiva que a gentamicina. A LAAO apresentou efeito citotóxico de aproximadamente 100% sobre as duas linhagens de células tumorais estudadas (B16F10 e MDA-MB-231), quando administrada na concentração de 1 μg/mL. Porém, nesta mesma concentração não apresentou nenhum efeito citotóxico sobre eritrócitos humanos. Desta forma, é possível inferir que os métodos empregados para o isolamento da LAAO foram eficientes e que se trata de uma enzima multifuncional com potencial uso terapêutico. Sugere-se o desenvolvimento de estudos voltados a esta temática com o intuito de ampliar a caracterização desta enzima e avaliar outras potenciais
ações bioativas. Snake venoms are rich sources of proteins, enzymes and peptides, which
represent 90% of the total constituents and probably are responsible for most of its biological effects. The L-amino acid oxidase (LAAO) is a component of venom and has antimicrobial, platelet aggregation-inducing, apoptoticinducing and anti-cancer activities. Considering the actions of the LAAOs, this work aims to characterize an isolated LAAO from crude venom of Bothrops jararacussu and evaluate its antibacterial and antitumor potential. The LAAO isolation involved three purification steps: an affinity chromatography on Con A-Sepharose column and two steps of ion exchange chromatography on MonoQ column. The protein purity was confirmed by SDS-PAGE and the obtained molecular mass was 68 kDa. The isolated LAAO showed higher affinity to L-Phe, followed by L-Met and L-Arg. The tryptic peptides profile revealed homology to other isolated LAAO from B. jararacussu, but the N-terminal sequence (ATNRNPLYYQFRRTDYFIF) displayed 56% identity with this enzyme, and therefore may be a new LAAO from B. jararacussu. The isolated LAAO showed antimicrobial activity against Gram-negative Proteus vulgaris (MIC 25 μg/mL, MBC 50 μg/mL) and Citrobacter freundii (MIC 100 μg/mL, MBC 100 μg/mL) and Gram-positive Listeria monocytogenes (MIC 25 μg/mL), Bacillus cereus (MIC 400 μg/mL), Bacillus subtilis (MIC 100 μg/mL, MBC 200 μg/mL) and Staphylococcus aureus (MIC 50 μg/mL, MBC 50 μg/mL), for which this enzyme was more effective than gentamicin. The LAAO displayed a cytotoxic effect of nearly 100% over two tumor cell lines studied (B16F10 and MDA-MB-231) when the enzyme was administrated at concentration 1 μg/mL. However the LAAO in
this same concentration did not show any cytotoxic effect over human
erythrocytes, which suggested that it s a multifunctional enzyme with therapeutic potential. The development of further studies involving this theme is necessary in order to enlarge enzyme characterization and to evaluate other potential bioactive actions.
