We report the first genetic analysis of archeological maize specimens from the site of San Lorenzo (1,500-700 BP) (Azapa valley, Arica, Chile). Ancient DNA was successfully isolated from 11 archeological maize grains. The Alcohol dehydrogenase 2 (Adh2) gene was analyzed because it has a highly variable region due to the presence of a microsatellite region around -28 to -8, which consists of GA repeats that may be present in three types GA(n), GA(n)TA and GA(1)AA(1)GA(n), which is used as an informative region of the routes of initial dispersion of maize. Five Adh2 alleles were obtained and the alignment of these sequences according to the variable region revealed the presence of the three types of GA repeated. Our results do not provide sufficient evidence to reject any maize spread model proposed. This is the first report focused on genetic analysis of maize associated with an archeological site in Chile. Reportamos el primer análisis genético de especímenes arqueológicos de maíz del sitio de San Lorenzo (1.500-700 AP) (valle de Azapa, Arica, Chile). El ADN antiguo se aisló con éxito de 11 granos de maíz arqueológicos. Se analizó el gen de la alcohol deshidrogenasa 2 (Adh2) porque tiene una región muy variable debido a la presencia de una región microsatélite alrededor de -28 a -8, que consiste en repeticiones GA que pueden estar presentes en tres tipos GA(n), GA (n)TA y GA(1)AA(1)GA(n), que se utiliza como región informativa de las rutas de dispersión inicial del maíz. Se obtuvieron cinco alelos Adh2 y el alineamiento de estas secuencias según la región variable reveló la presencia de los tres tipos de GA repetidos. Nuestros resultados no proporcionan evidencia suficiente para rechazar cualquier modelo de dispersión de maíz propuesto. Este es el primer reporte enfocado en el análisis genético de maíz asociado a un sitio arqueológico en Chile.
