dc.description | The cerebellum plays a key role in motor tasks, but its involvement in cognition is still being considered. Although there is an association of different psychiatric and cognitive disorders with cerebellar impairments, the lack of time-course studies has hindered the understanding of the involvement of cerebellum in cognitive and non-motor functions. Such association was here studied using the Purkinje Cell Degeneration mutant mouse, a model of selective and progressive cerebellar degeneration that lacks the cytosolic carboxypeptidase 1 (CCP1). The effects of the absence of this enzyme on the cerebellum of mutant mice were analyzed both in vitro and in vivo. These analyses were carried out longitudinally (throughout both the pre-neurodegenerative and neurodegenerative stages) and different motor and non-motor tests were performed. We demonstrate that the lack of CCP1 affects microtubule dynamics and flexibility, defects that contribute to the morphological alterations of the Purkinje cells (PCs), and to progressive cerebellar breakdown. Moreover, this degeneration led not only to motor defects but also to gradual cognitive impairments, directly related to the progression of cellular damage. Our findings confirm the cerebellar implication in non-motor tasks, where the formation of the healthy, typical PCs structure is necessary for normal cognitive and affective behavior. | |
dc.description | El cerebelo juega un papel clave en las tareas motoras, pero aún se está considerando su participación en la cognición. Aunque existe una asociación de diferentes trastornos psiquiátricos y cognitivos con alteraciones del cerebelo, la falta de estudios a lo largo del tiempo ha dificultado la comprensión de la participación del cerebelo en las funciones cognitivas y no motoras. Dicha asociación se estudió aquí utilizando el ratón mutante Purkinje Cell Degeneration, un modelo de degeneración cerebelosa selectiva y progresiva que carece de la carboxipeptidasa 1 citosólica (CCP1). Los efectos de la ausencia de esta enzima sobre el cerebelo de ratones mutantes se analizaron tanto in vitro como in vivo. Estos análisis se realizaron de forma longitudinal (tanto en la etapa preneurodegenerativa como en la neurodegenerativa) y se realizaron diferentes pruebas motoras y no motoras. Demostramos que la falta de CCP1 afecta la dinámica y la flexibilidad de los microtúbulos, defectos que contribuyen a las alteraciones morfológicas de las células de Purkinje (PC) y al deterioro progresivo del cerebelo. Además, esta degeneración condujo no solo a defectos motores sino también a deterioros cognitivos graduales, directamente relacionados con la progresión del daño celular. Nuestros hallazgos confirman la implicación del cerebelo en tareas no motoras, donde la formación de la estructura típica y saludable de las PC es necesaria para el comportamiento cognitivo y afectivo normal. | |
