Background and Purpose: Study aiming to investigate the effect of nanostructured
curcumin microemulsion in the treatment of experimental ulcerative colitis (UC) in rats.
Methods: Colitis was induced by intracolonic instillation of 4% acetic acid (AAC). The
animals were treated for seven days and randomly assigned to four groups: NC
(control), colitis/control (AAC+SN), colitis/curcumin (AAC+CUR) and colitis/mesalazine
(AAC+MES). Dosage of inflammatory cytokines and antioxidant enzymes, as well as
macro and microscopic evaluation of the colons were analyzed. Results: The group
treated with curcumin (AAC+CUR) had higher dosage of antioxidant enzymes, when
compared to the AAC+MES group. There was a significant reduction in the levels of
the pro-inflammatory cytokines TNF- α , IL-1 β and IL-6 in the colonic tissue of the
animals in the AAC+CUR group, when compared with the AAC+SN and AAC+MES
groups (p<0.001 ). The presence of mucosal ulcers (MU) in the group treated
exclusively with acetic acid (AAC+SN) was significantly higher (p=0.001), compared to
the mesalazine group (AAC+MES). In the control and curcumin-treated groups
(AAC+CUR) there was no presence of ulcers in the colonic mucosa. Conclusion:
Curcumin, used orally, in the nanostructured microemulsion formulation, positively
influences the treatment of experimental colitis in rats. The results also suggest that
nanostructured curcumin with negative electrical charge is a promising
phytopharmaceutical delivery system for UC therapy. Future studies are needed to
better clarify the mechanisms of the effects of nanostructured curcumin microemulsion
in the treatment of UC. Introdução/Objetivo: Estudo com o objetivo de investigar o efeito de microemulsão
nanoestruturada de curcumina com potencial zeta negativo no tratamento da colite
ulcerativa (UC) experimental em ratos. Métodos: A colite foi induzida por instilação
intra-colônica de ácido acético 4% (AAC). Os animais foram tratados por sete dias e
distribuídos randomicamente em quatro grupos: CN (controle), colite/controle
(AAC+SN), colite/curcumina (AAC+CUR) e colite/mesalazina (AAC+MES). Foram
analisados parâmetros clínicos, dosagem de citocinas inflamatórias e de enzimas
antioxidantes, além de avaliação macro e microscópica dos cólons. Resultados: O
grupo tratado com curcumina (AAC+CUR) obteve maior dosagem de enzimas
antioxidantes, quando comparado com o grupo AAC+MES. Houve uma redução
significante dos níveis das citocinas pró-inflamatórias TNF-α, IL-1β e IL-6 no tecido
colônico dos animais do grupo AAC+CUR, quando comparado com os grupos
AAC+SN e AAC+MES (p<0,001). A presença de úlceras de mucosa (UM) no grupo
tratado exclusivamente com ácido acético (AAC+SN) foi significativamente maior
(p=0,001), comparando-se com o grupo mesalazina (AAC+MES). Nos grupos controle
e tratado com curcumina (AAC+CUR) não houve presença de úlceras na mucosa
colônica. Conclusão: A curcumina, usada por via oral, na formulação microemulsão
nanoestruturada, influencia positivamente no tratamento da colite experimental em
ratos. Os resultados também sugerem que a curcumina nanoestruturada com
potencial de carga elétrica negativa é um sistema de liberação de fitofármaco
promissor para a terapia de UC. Estudos futuros são necessários para melhor
esclarecer os mecanismos dos efeitos da microemulsão nanoestruturada de
curcumina no tratamento da UC.
