RESUMEN: El presente documento tiene como propósito dar a conocer los resultados obtenidos en el trabajo de investigación: “Variabilidad fenotípica y genética de Tithonia diversifolia (hemsl.) A. Gray, una planta para la producción animal sostenible en Colombia”, desarrollado para optar por el título de Doctor en Ciencias Animales.
Con el fin de agrupar los resultados obtenidos en cada una de las temáticas trabajadas, este documento de tesis esta conformado por seis capítulos estructurados en forma de artículos científicos y su objetivo general es evaluar diferentes genotipos de T. diversifolia e identificar aquellos con mayor potencial para la producción forrajera en sistemas con rumiantes. En el primer capítulo se revisa la literatura sobre la diversidad química y agronómica de T. diversifolia y sus usos en la alimentación animal y en otras áreas; el capítulo dos reporta la diversidad genética de esta arbustiva en materiales colectados en Colombia y México, y su potencial para la alimentación de rumiantes; el tercer capítulo evalúa la interacción GxA y la estabilidad de genotipos destacados de T. diversifolia para la producción de biomasa y su composición química bajo diferentes condiciones edafoclimáticas; el cuarto capítulo muestra los resultados de una evaluación in vitro de la dinámica de fermentación y una cuantificación de AGV y CH4 de dietas con la inclusión de diferentes genotipos de T. diversifolia; en el quinto capítulo se describen las fases de crecimiento, desarrollo y reproducción de esta especie junto con su potencial de producción de semilla sexual y su germinación; y finalmente en el sexto capítulo se reporta una evaluación del consumo voluntario y selectividad de los diferentes genotipos estudiados por bovinos en fase de crecimiento.
La revisión de literatura se basó en una búsqueda de artículos publicados en revistas científicas durante los últimos 10 años. Se encontró que, desde el punto de vista fitoquímico se han identificado más de 150 compuestos presentes en T. diversifolia, algunos con propiedades farmacológicas o capaces de modular la dinámica de degradación y fermentación en el rumen. También se destaca que T. diversifolia tiene una alta adaptabilidad a condiciones edafoclimáticas adversas lo que puede atribuirse en parte a la capacidad de asociarse con microorganismos diazotróficos del suelo y solubilizadores de fosfatos, además, de su diversidad y plasticidad genotípica y fenotípica.
En cuanto al análisis diversidad genética (capítulo Dos), se evaluaron 31 poblaciones seleccionadas y colectadas en Colombia y México. El análisis de diversidad genética se realizó mediante marcadores moleculares ISSR y del gen del citocromo P450 y sus características químicas y morfologías fueron analizadas con estadística multivariada. Los materiales evaluados presentaron diferencias en el contenido de PC (29.4% ± 3.29), EE (1.6% ± 0.67), Ca (2.3% ± 0.2), P (0.6% ± 0.18), FDA (44.99 ± 9.43) y FDN (46.82 ± 12.52). Igualmente, se encontraron diferencias en el peso total de la planta, área foliar, hojas por rama y altura de planta (p<0.05). Finalmente se identificó una amplia diversidad genética con cinco grupos bien definidos y mediana relación geográfica entre materiales, con un índice de Nei de 0.281 y un índice de Shannon de 0.432.
La interacción GxA se evaluó en siete genotipos destacados para la producción de MS en tres ambientes y se midieron algunas características químicas y morfológicas. La interacción GxA se estudió con los modelos AMMI y SREG. Para la producción de MS se encontraron diferencias significativas entre los genotipos (p < 0.05), en el análisis GxA la interacción resultó significativa y se observaron efectos del ambiente sobre la productividad (p<0.05). De otro lado, fueron encontrados dos genotipos estables y otros con mayor adaptación a cada uno de los ambientales.
En el estudio de la dinámica de fermentación y degradación ruminal, se evaluaron mezclas de siete materiales de T. diversifolia y las gramíneas Cenchrus clandestinus y Urochloa brizantha cv Marandú. La inclusión de T. diversifolia en dietas basadas en U. brizantha mejoró la oferta de nutrientes y modificó los parámetros de dinámica de fermentación (p<0.05); la degradación de la MS se incrementó 4.72 puntos porcentuales frente a dietas únicamente con esta gramínea (p<0.05), las emisiones de CH4 fueron significativamente menores en dietas que tuvieron T. diversifolia (p<0.05) y se disminuyó la relación A:P. En dietas basadas en C. clandestinus, la inclusión de T. diversifolia modificó la composición química de las dietas y la degradación de la MS (p<0.05), pero tuvo poco efecto en los parámetros de dinámica de fermentación y en sus productos.
En la evaluación de los aspectos reproductivos los genotipos tuvieron diferencias significativas en todas las variables medidas a excepción del tiempo de secado de aquenios y del porcentaje de semillas rudimentarias (p<0.05). En la producción de semilla sexual, la fertilización tuvo efecto significado (p< 0.05) y en general incrementó el tiempo de las fases evaluadas; con relación al porcentaje de germinación hubo diferencias significativas entre genotipos y entre los tratamientos (p =0.000118), y se encontró que algunos procesos pre-germinativos pueden incrementar el porcentaje de germinación.
Finalmente, en las determinaciones de consumo se encontró una ingesta de 0.39 ± 0.045 kg de MS en cada periodo de evaluación con diferencias significativas entre algunos genotipos, pero todos los materiales fueron consumidos. No se encontró diferencias en cuanto al tiempo en el comedero entre los genotipos (p=0.0818).
Se concluye que T. diversifolia es una especie que puede ser utilizada como arbustiva forrajera bajo diferentes condiciones edafoclimáticas como suelos ácidos y de baja fertilidad gracias a su alta diversidad genética. A pesar de que se identificó que esta especie ofrece una alta calidad nutricional debido a su baja fibra, alta oferta de nutrientes y degradabilidad existe la posibilidad de seleccionar materiales superiores para el crecimiento y producción de biomasa para condiciones específicas. "Phenotypic and genetic variability of Tithonia diversifolia (hemsl.) A. Gray, a plant for
sustainable animal production in Colombia", carried out to opt for the title of PhD in Animal
Science.
In order to group the results obtained around specific topics, this thesis document is
comprised of six chapters structured in the form of scientific articles and its general
objective is to evaluate different genotypes of T. diversifolia and identify those with the
greatest potential for forage production in systems with ruminants. The first chapter
reviews the literature on the chemical and agronomic diversity of T. diversifolia and its
uses in animal feed and in other areas; Chapter two reports on the genetic diversity of this
shrub in accessions collected in Colombia and Mexico, and its potential for feeding
ruminants; the third chapter reports the GxA interaction and the stability of prominent T.
diversifolia genotypes for biomass production and its chemical composition under different
edaphoclimatic conditions; the fourth chapter reports the results of an evaluation of the
fermentation dynamics and a quantification of AGV and CH4 from diets with the inclusion
of different genotypes of T. diversifolia; in the fifth chapter the growth, development and
reproduction phases of this species are described together with its potential for the
production of sexual seed and its germination; and finally in the sixth chapter an evaluation
of the voluntary consumption of the different genotypes studied by growing bovines is
reported.
The review was based on a search for articles published in scientific journals during the
last 10 years. It was found that T. diversifolia has more than 150 compounds, some with
pharmacological properties or capable of modulating the dynamics of degradation and
fermentation in the rumen. It is also highlighted that T. diversifolia has a high adaptability
to adverse edaphoclimatic conditions, which can be partially attributed to its ability to
associate with diazotrophic and phosphate solubilizer microorganisms in the soil, in
addition to its diversity and genotypic and phenotypic plasticity.
Regarding the genetic diversity analysis (Chapter Two), 31 selected and collected
populations in Colombia and Mexico were evaluated. Genetic diversity analysis was
performed using ISSR molecular markers and the cytochrome P450 gene, and their
chemical characteristics and morphologies were analyzed with multivariate statistics. The
genotypes evaluated showed differences in the CP content (29.4% ± 3.29), EE (1.6% ±
0.67), Ca (2.3% ± 0.2), P (0.6% ± 0.18), ADF (44.99 ± 9.43) and NDF (46.82 ± 12.52) (p
<0.05). Also, differences were found in the total weight of the plant, leaf area, leaves per
branch and plant height (p <0.05). Finally, a high genetic diversity was identified with five
defined groups and a medium geographic relationship between materials, with a Nei index
of 0.281 and a Shannon index of 0.432. The GxE interaction included seven genotypes evaluated for the production of DM in three
environments and also some chemical and morphological characteristics were measured.
The GxE interaction was studied with the AMMI and SREG models. For the production of
DM, significant differences were found between genotypes (p <0.05), in the GxE analysis
the interaction was significant and effects of the environment on productivity were
observed (p <0.05). On the other hand, two stable genotypes were identified in addition
to materials with greater adaptation capacity to each of the environments.
In the study of the fermentation and ruminal degradation, mixtures of seven materials of
T. diversifolia, with Cenchrus clandestinus and Urochloa brizantha cv Marandú grasses
were evaluated. Inclusion of T. diversifolia in diets based on U. brizantha improved the
supply of nutrients (less fiber, more protein, fat and minerals) and modified the
fermentation parameters (p<0.05); the DM degradation increased 4.72% compared to
diets with only this grass (p <0.05) and CH4 emissions were significantly lower in diets that
had T. diversifolia (p <0.05). Finally, in diets with C. clandestinus, the inclusion of T.
diversifolia modified the chemical composition in the diets and the degradation of DM (p
<0.05) but did not affect fermentation parameters.
In the reproductive evaluation, the genotypes had significant differences in all the
variables measured except for the drying time of achenes and percentage of rudimentary
seeds (p <0.05). In sexual seed production, fertilization had a significant effect (p <0.05)
and in general increased the time of the phases evaluated; and regarding the germination
percentage there were significant differences between genotypes and between
treatments (p = 0.000118), with some pre-germination processes increasing the
germination percentage.
Finally, observed biomass intake was 0.39 ± 0.045 kg of DM per day, with significant
differences between some genotypes, but all materials were consumed. In intake time
there were no differences between genotypes (p = 0.0818).
It is concluded that T. diversifolia is a species that can be used as a forage shrub under
different edaphoclimatic conditions due to its high genetic diversity. Although it was
identified that this species offers nutritional quality, there is the possibility of selecting
superior materials for biomass growth and production.
