Metabolic and functional analysis of an aquous extract of Ugni molinae Turcz on refrigerated boar sperm.

The world pork market grows every year. This type of food is a good source of B vitamins and is considered a cheap alternative of animal protein. In the pig breeding industry, artificial insemination (AI) is the best system to increase the number of animals and decrease the interval between generations, a combination of factors that improves breeding programs, in this sense the refrigeration of swine semen is the alternative currently used by the pig farms to perform the artificial insemination, a system based on refrigeration at 17 ° C of the seminal dose in commercial extenders. This refrigeration alters the biochemical and physiological characteristics of the cells, especially by the endogenous production of the reactive oxygen species (ROS) that are mainly produced by the destabilization of the mitochondria. In this sense, antioxidants play a fundamental role in the regulation of oxidative imbalance. In this study, a pharmacological analysis of an extract rich in polyphenols of the fruit of murtilla (Ugni molinae Turcz.) That has a high content of antioxidant agents was carried out. We evaluated the potential regulatory effect of oxidative imbalance in sperm cells stimulated with hydrogen peroxide (H2O2) as an oxidizing agent to determine the antioxidant potential in the different cellular phases, characterization of effective concentrations for the regulation of this REDOX state and evidencing direct effects on the most important physiological and biochemical components in sperm activation, intracellular calcium movement, cAMP, and processes associated with ATP by the aerobic pathway. The results suggest a substantial antioxidant effect that focuses mainly on the reduction of extracellular and intracellular ROS but to a lesser extent the regulation of lipid peroxidation of the membrane. This effect was observed with the complete extract and with the main flavonoids detected in the extract, which may have synergistic, additive effects as well as antagonistic effects. On the other hand, it was observed that the supplementation of the commercial diluent with murtilla, implies in a reduction of the ATP content, mediated by the decrease of the mitochondrial membrane potential, diminution of the content of cAMP and intracellular calcium movement, which implies an effect direct in sperm capacitation, however, the regulatory effect of ROS and mitochondrial potential / oxygen consumption, reduce the impact of this imbalance in the long term, improving the motility and ROS indicators up to 196 hours of refrigeration. These results allow us to conclude that the extract and / or its main flavonoids such as quercetin and catechin can be used as antioxidant supplements to extend the useful life of the seminal dose.
