Uso de Nanopartículas luminiscentes para estudiarlos efectos químico y térmico por radiación en células

Resumen
La interacción de la radiación con el tejido biológico produce, al menos, dos tipos de efectos: químico y térmico. El efecto químico es asociado a la formación de especies reactivas de oxigeno (ROS) generadas por ionización, mientras que el térmico consiste en el incremento de la temperatura debido a la absorción de energía térmica inducida por un láser. En este trabajo se estudiaron los fenómenos químico y térmico inducido por radiación en células asistido con nanopartículas (NPs). El efecto químico se indujo irradiando la sangre con una fuente de Cobalto 60. La oxigenación celular se correlacionó a través de espectroscopia Raman con la ayuda de nanodiamantes carboxilados (cNDs), y las alteraciones morfológicas se evaluaron con microscopia de fuerza atómica (AFM). Por su parte, el efecto térmico se indujo atrapando ópticamente las células con láseres de 820 y 980 nm aplicando potencias de 20 a 250 mW, se utilizaron NPs de CdSe para determinar la temperatura generada, y de igual forma se analizaron los efectos. Nuestros resultados muestran que, independientemente de la naturaleza de la radiación, la membrana celular se compromete cambiando sus propiedades fisicoquímicas y estructurales. Se demostró la eficiencia de los cNDs como nanodetectores de los estados de oxigenación, y de los QDs como nanosensores de temperatura sin afectar las condiciones experimentales ni la viabilidad celular.
