RESUMEN: La obesidad es un problema de salud pública a nivel mundial. Esta enfermedad se ha asociado al consumo de alimentos con alta densidad energética basados en un contenido alto de grasas y azúcares refinados; así como una disminución del consumo de alimentos con fibra dietética. Las agavinas (AV) (Agave angustifolia Haw) y la fracción indigerible (FI) de la harina de plátano verde (HPV) (Musa paradisiaca L.) son consideradas como fibra dietética. Estos compuestos han sido
estudiados por diversos grupos de investigación como ingredientes nutracéuticos. El objetivo de este trabajo fue evaluar la fermentación colónica in vitro de la mezcla de AV y la FI de la harina de plátano verde en una relación 1:1 p/p. Así como también, evaluar el efecto del consumo de la combinación de AV + HPV en cambios metabólicos, morfológicos, inflamación sistémica y permeabilidad intestinal en un modelo de obesidad en murinos. En la fermentación colónica in vitro se identificó el perfil de AGCC mediante cromatografía (CG), y se observó que la generación total de AGCC fue
significativamente mayor por efecto de la mezcla comparado con AV y HPV de manera individual. En el modelo de obesidad en murinos se observó que el consumo la dieta alta en grasa (AG) modificó significativamente el metabolismo de los ratones aumentando el contenido de glucosa, triglicéridos y presión arterial. Sin embargo, el consumo de la dieta AG suplementada con la mezcla (AV:HPV), disminuyó los parámetros bioquímicos evaluados en los ratones. El consumo de la dieta AG también
indujo un grado de inflamación crónica y cambios en el tejido del colon de los ratones. Pero, el consumo de la dieta AG suplementada con la mezcla (AV:HPV) disminuyó la inflamación crónica evaluado mediante marcadores de inflamación como el TNF-α, IL-1 y la IL-6. Además, se observó mediante técnicas de histología, el cambio en el tejido del colon por efecto del consumo de la dieta AG, el cual disminuyó significativamente con la suplementación de la mezcla (AV:HPV). Estos resultados mostraron que la mezcla de fibra dietética con diferente grado de fermentabilidad y composición, es una alternativa que complementa los efectos benéficos del consumo de fibra dietética y la modulación de la microbiota intestinal.
ABSTRACT: Obesity is a public health problem worldwide. This disease has been associated with the consumption of high energy density food, based on a high content of fats and refined sugars, as well as a decrease in the consumption of dietary fiber. Agavins (AV) (Agave angustifolia Haw) and the indigestible fraction (IF) of green banana flour (GBF) (Musa paradisiaca L.) are considered as dietary fiber. These compounds have studied as nutraceutical ingredients. The objective of this work was to evaluate the profile of shortchain fatty acids (SCFA) through in vitro colonic fermentation of the mixture of AV and IF of GBF in a 1: 1 ratio p/p. To evaluate the effect of the consumption of the combination of AV + GBF in metabolic, morphological, systemic inflammation, and intestinal permeability in a mouse model of obesity. In the in vitro colonic fermentation, the SCFA profile was identified by a gas chromatograph (GC), it was observed that the total generation of SCFA was significantly higher due to the effect of the mixture compared with AV and GBF individually. In a mouse model of obesity, it was observed that the consumption of the high-fat diet (HF) significantly modified the metabolism of mice by increasing the content of glucose, triglycerides, and blood pressure. However, the consumption of the HF-diet supplemented with the mixture (AV: GBF), decreased the biochemical parameters evaluated in mice fed with this diet. The consumption of HF-diet induced a degree of chronic inflammation and changes in the colon tissue of the mice. Nevertheless, consumption of the AG diet supplemented with the mixture (AV: GBF) decreased chronic inflammation assessed by inflammation markers such as TNF-α, IL-1, and IL-6. The change in colon tissue due to the consumption of an HF-diet, which was significantly reduced by the consumption of the mixture (AV: GBF), was observed by histology techniques. These results demonstrate that the mixture of dietary fiber with different degrees of fermentability and composition is an alternative that complements its effects and modulated of the intestinal microbiota.
